不同频率压力刺激对兔软骨细胞整合素α1β1的影响

Effects of Different Frequency Pressures on Integrin-α1β1 in Rabbit Chondrocytes

摘要 目的:观察不同频率压力对兔关节软骨细胞增殖及其分泌整合素α1β1的影响。方法:通过体外分离培养胎兔关节软骨细胞,取第3代细胞接种于6孔板,分别设置无干预对照组(A组)、不同频率的加压组(压力25PSI,频率:B1组0.2Hz、B2组0.5Hz、B3组1Hz、B4组2Hz、B5组5Hz、B6组持续加压)。压力干预每天2h,连续干预3d后,以WST-1细胞增殖及细胞毒性检测试剂盒检测各组软骨细胞增殖情况(OD值),并以ELISA法检测各组培养上清中整合素α1β1含量。结果:细胞OD值A组为1.08±0.13,B1组为1.19±0.11,B2组为1.23±0.08,B3组为1.35±0.06,B4组为1.45±0.09,B5组为1.37±0.12,B6组为1.21±0.12。加压各组与对照组比较OD值有升高,差异有统计学意义。B组各组间差异有统计学意义,峰值出现在B4组。A组整合素α1β1水平为(3.02±0.26)pg/mL,B1组为(3.28±0.19)pg/mL,B2组为(3.43±0.11)pg/mL,B3组为(3.77±0.24)pg/mL,B4组为(4.01±0.09)pg/mL,B5组为(3.94±0.13)pg/mL,B6组为(3.37±0.13)pg/mL。加压各组整合素α1β1值与对照组比较有所升高(P<0.05)。B组各组间比较差异有统计学意义,峰值出现在B4组。结论:相同大小(25PSI)不同频率的压力刺激兔软骨细胞,均能增加软骨细胞的增殖作用和整合素的含量。其中以2Hz的频率作用最强。 Objective:To observe the effect of different frequency pressures on proliferation of rabbit articular chondrocytes and integrin-α1β1production.Methods:The chondrocytes were isolated form fetal rabbit articular cartilage and cultured in vitro;third-generation chondrocytes were plated in culture plates,and the normal pressure was 25 PSI,and the frequency of every group was different(B2group was 0.5Hz,B3 group was 1Hz,B4 group was 2Hz,B5 group was 5Hz,B6 group received continuous pressure).The pressure lasted 2hours a day.After continuous intervention for 72 hours,the cell absorbance(optical density)was evaluated by WST-1Cell Proliferation and Cytotoxicity Assay Kit.The concentration of integrin-α1β1in supernatant of each group was detected by ELISA.Results:There were significant differences in OD value and integrin-α1β1concentration between the A group and all other groups(P〈0.05).There was significant difference in integrin-α1β1concentration among all the groups(B1to B6,P〈0.05).Conclusion:The same pressure(25PSI)stimulation under different frequencies can increase the cartilage cell proliferation and the content of integrin,and the 2Hz frequency was strongest.
