大肠埃希菌耐药基因及可移动遗传元件与噬菌体原遗传标记研究 被引量:4

摘要 目的调查大肠埃希菌临床分离株的耐药性及遗传学特征。方法收集2009年1-12月患者尿液标本中分离的大肠埃希菌20株,采用聚合酶链反应(PCR)及序列分析的方法分析23种β-内酰胺酶基因、14种氨基糖苷类修饰酶基因、6种16SrRNA甲基化酶基因、8种可移动遗传元件遗传标记、9种噬菌体原遗传标记以及与细菌细胞分裂相关的2种基因(dicB、kilR),与细菌致病性相关的2种基因(hofQ、ompT)。结果 20株大肠埃希菌共检出3种β-内酰胺酶基因、6种氨基糖苷类修饰酶基因、1种16SrRNA甲基化酶基因,7种可移动遗传元件遗传标记、6种噬菌体原遗传标记、2种与细菌细胞分裂相关的基因、2种与细菌致病性相关的基因;20株大肠埃希菌全部基因检测结果的样本聚类分析示,1、4、9、10号株为一簇,其余为另一簇。结论对大肠埃希菌尿液分离株进行β-内酰胺酶基因、氨基糖苷类修饰酶基因、16SrRNA甲基化酶基因、可移动遗传元件遗传标记、噬菌体原遗传标记以及与细菌细胞分裂相关的基因,与细菌致病性相关的基因检测与分析,国内为首次,从样本聚类分析图可见,虽无克隆传播,但6、12号株与7、18号株有较近的亲缘关系,有医院感染的可能性。 OBJECTIVE To investigate the resistance and genetic characteristics of a group of 20 strains of Esche-richia coli .METHODS Totally 20 strains of E. coli isolated from urine in patients from Jan .2009 to Dec .2009 were collected .Then, 23 kinds of beta-lactamase genes, 14 kinds of aminoglycoside modifying enzyme genes, 6 kinds of 16S rRNA methylase genes, 8 kinds of mobile genetic elements, 9 kinds of prophages, 2 kinds of cell division genes (dicB, kilR), 2 kinds of pathogenicity genes (hofQ, ompT) were analyzed by PCR and verified by DNA sequencing .RESULTS In 20 strains of E. coli, 3 kinds of beta-lactamase genes, 6 kinds of aminoglycoside modifying enzyme genes, 1 kind of 16SrRNA methylase genes, 7 kinds of mobile genetic elements, 6 kinds of prophages, 2 kinds of cell division genes, 2 kinds of pathogenicity genes were positive .Sample cluster analysis showed that strains No .1, 4 and strains No .9, 10 were in the same cluster, other strains were in the other cluster .CONCLUSIONS It′s the first report to detect beta-lactamase genes, aminoglycoside modifying enzyme genes, 16S rRNA methylase genes, mobile genetic elements, prophages, cell division genes, pathogenicity genes in China .Sample cluster analysis showed that strains No .6, 12 and strains No .7, 18 were close in relationship, and might cause nosocomial infection.
