病毒灭活血浆对人自然杀伤细胞功能影响的体外实验研究

摘要 目的:探讨病毒灭活血浆对人自然杀伤细胞( natural killer cell ,NK细胞)生长、细胞表型以及体外杀伤活性的影响。方法采用病毒灭活血浆和新鲜冰冻血浆分别培养人NK细胞,观察2组培养体系中NK细胞的扩增情况;用流式细胞仪(FCM)检测NK细胞CD3-CD56+表达以及NK细胞穿孔素、颗粒酶B和CD107a的含量变化;用乳酸脱氢酶法测定NK细胞杀伤SGC-7901细胞活性。结果病毒灭活血浆与新鲜冰冻血浆比较,在促进NK细胞的增殖以及CD3-CD56+的表达上无明显差异( P>0.05);检测培养10天的NK细胞,病毒灭活血浆培养组细胞穿孔素、颗粒酶B和CD107a以及杀伤活性与新鲜冰冻血浆培养组也无明显差异(P>0.05)。结论病毒灭活血浆对人NK细胞的增殖、细胞表型和体外杀伤功能均无明显影响。 Objective To investigate the effect of virus inactivated plasma on human NK cell growth , cell phenotype and in vitro cytotoxicity.Methods The virus inactivated plasma and fresh frozen plasma were used to culture NK cells respectively , for the purpose to compare the two kinds of culture systems on NK cell amplification .The expression of CD3 +CD56 +, perforin and granzyme B on NK cells were detected by flow cytometry (FCM).LDH assay was used to measure cytotoxic activity of NK cells .Results Fresh frozen plasma and virus inactivated fresh frozen plasma were not significantly different on NK cell growth, phenotype expression and cytotoxicity (P〉0.05).Detection of NK cell cul-tured with fresh frozen plasma and virus inactivated fresh frozen plasma for 10 days showed no significant difference in the expression of CD3 +CD56 +, perforin and granzyme B (P〉0.05).Conclusion The virus inactivated plasma has no effect on proliferation , expression and function of human NK cells in vitro.
