不同物种支原体的比较基因组学研究

摘要 为了研究不同物种支原体在进化关系、结构组成以及致病因子三方面所存在的异同,试验采用比较基因组学的方法,分析了不同物种来源的21株支原体的进化关系,并以肺炎支原体为例,对来自三个不同物种(猪、牛、羊)的代表株在基因组的组成结构以及整体相似程度方面进行了横向比较分析,并进一步利用相似性比较对这三个物种代表株进行潜在的毒力因子挖掘。结果表明:三个不同物种的肺炎支原体差异较大;猪肺炎支原体内毒力因子的保守性和特异性都较高,牛支原体和山羊支原体中存在较多相似的毒力因子。说明以不同宿主寄生的致病型支原体结构组成上变异较大,但羊与牛的肺炎支原体在致病机理上可能更为相近。 To study the similarities and differences between Mycoplasma strains in the evolutionary relationships, structure and composition, and virulence factors, a method of comparative genomics was used to analyze the evolutionary relationships among 21 strains of Mycoplasma from dif- ferent species. Mycoplasma pneumoniae was used as an example, and the genome structure and composition with overall similarity of the repre- sentative strains of Mycoplasma pneumoniae from the three species (pig, cattle, and goat) were compared and analyzed horizontally, and moreo- ver, their potential virulence factors were explored further by similarity alignment. The results showed that the representative strains of Mycoplas- ma pneumoniae from the three species were quite different ; the conservation and specificity of the virulence factors of Mycoplasma hyopneumoni- ae were higher than those of the others ; there were more similar virulence factors between Mycoplasma bovis and Mycoplasma capricolum. The re- suits indicate that the pathogenic mycoplasmas from different hosts were different on the genome structure and composition, but Mycoplasma bovis may be similar to Mycoplasma capricolum in pathogenic mechanism.
