北京鸭和淮南麻鸭胸肌发育过程TPM1和TPM3基因的表达变化研究 被引量:5

Study on the changes in the TPM1 and TPM3 gene expressions during the development of breast muscle in Peking duck and Huainan shelduck

摘要 为了解北京鸭和淮南麻鸭胸肌发育过程TPM1和TPM3基因的表达变化,试验分别采集出壳当天(1 d)、2周龄、4周龄、6周龄和8周龄北京鸭和淮南麻鸭胸肌组织,提取总RNA,利用qRTPCR技术分析TPM1和TPM3基因在北京鸭和淮南麻鸭胸肌发育过程的表达变化情况。结果表明:TPM1基因在北京鸭和淮南麻鸭胸肌发育过程中没有明显的变化(P>0.05)。TPM3基因在出壳当天北京鸭和淮南麻鸭胸肌组织中的表达量与其他时间点相比显著提高(P<0.05),在2周龄淮南麻鸭胸肌组织中的表达量与6周龄和8周龄相比显著降低(P<0.05),在4周龄淮南麻鸭胸肌组织中的表达量没有显著变化(P>0.05)。从出壳当天到8周龄时,TPM3基因在北京鸭胸肌组织中的表达量与淮南麻鸭胸肌组织中的表达量相比有升高的趋势。 To understand the changes in the TPM1 and TPM3 gene expressions during the development of breast muscle in Peking duck and Huainan shelduck, the breast muscle tissue samples from Peking duck and Huainan shelduck were collected at the hatching day (1 d) ,2 weeks of age,4 weeks of age,6 weeks of age,and 8 weeks of age respectively. The total RNA was extracted,and then the changes in the the TPM1 and TPM3 gene expressions during the development of breast muscle in Peking duck and Huainan shelduck were analyzed by qRT - PCR technolo- gy. The results showed that no significant change was found in the TPM1 gene expressions during the breast development in Peking ducks and Huainan shelducks(P 〉0.05 ). The expression levels of the TPM3 gene from the breast muscle of Peking duck and Huainan shelduck at the hatching day were significantly increased ( P 〈 0.05 ) compared with those at other time points,while the expression level of the TPM3 gene from the breast muscle of Huainan shelduck at 2 weeks of age was significantly lower ( P 〈 0.05 ) than that at 6 and 8 weeks of age, but no significant changes ( P 〉 0.05 ) were found at in the expression level of the TPM3 gene from the breast muscle of Huainan shelduck at 4 weeks of age. The expression levels of the TPM3 gene from the breast muscle of Peking duck were higher than those of Huainan shelduck from the hatching day to 8 weeks of age.
