摘要 【目的】以含量测定为重点,开展奶牛隐性乳房炎诊断液的质量标准研究,为规范诊断液的技术标准提供参考。【方法】建立直接两相滴定法测定奶牛隐性乳房炎诊断液中的阴离子活性物含量,并以诊断液LMT为例,进行方法的准确度、精密度和专属性评价。【结果】准确度分析结果显示,RE为-0.86%(-3.88%-3.06%);精密度分析结果显示,日内RSD为0.57%-3.14%,日间RSD为2.19%;专属性分析结果显示,LMT中的辅料成分不影响阴离子活性物的测定。直接两相滴定法测定LMT中的阴离子活性物,具备必要的准确度、精密度和专属性。【结论】因为无论是组成还是技术,LMT均能代表同类的诊断液,所以直接两相滴定法可作为奶牛隐性乳房炎诊断液的含量测定方法。 Objective The quality standard of diagnostic reagent for subclinical mastitis in dairy cow was studied,with particular emphasis on content determination in order to regulate the technology standard of the diagnostic reagents. Method Direct two-phase titration method was developed to determinate the anionic active matter in diagnostic reagents for subclinical mastitis in dairy cow. Taking the diagnostic reagent LMT as an example,its accuracy,precision and specificity were evaluated. Result Accuracy analysis showed that the RE was -0.86%(-3.88-3.06%). Precision analysis showed that intra-day RSD and inter-day RSD were 0.57-3.14%and 2.19%respectively. Specificity analysis showed that the result was not affected by other accessory ingredients in LMT. Direct two-phase titration method was developed to determine anionic active matter in LMT with necessary accuracy,precision and specificity. Conclusion Because LMT is representative both in composition and technology in diagnostic reagents for subclinical mastitis in dairy cows,so direct two-phase titration method could be used to content determination of the diagnostic reagents.
