摘要
目的 研究热休克恢复期小鼠大脑、垂体、肾、肾上腺中HSP70的表达规律。 方法 昆明系小鼠随机分为对照组和实验组。实验组动物 :1 分别以 40℃ ,42℃ ,44℃ ,46℃热休克处理 30min ,恢复 2h和 8h ;2 以46℃热休克处理 30min ,恢复 2、4、6、8、12、2 4、48、72h ,以免疫组织化学结合图像分析技术观察HSP70的表达。 结果 1 42℃ ,44℃ ,46℃均能诱导肾、肾上腺表达HSP70 ,44℃、46℃可诱导大脑、垂体表达HSP70 ,但均以 46℃组阳性免疫反应面积最大 (P <0 0 1) ;2 HSP70的合成高峰在大脑和垂体为热休克恢复期 4~ 8h(P <0 0 1) ,肾和肾上腺为 8~ 12h(P <0 0 1) ;3 HSP70阳性免疫反应定位于大脑神经膜和郎飞结 ,垂体神经膜 ,肾小管、肾上腺网状带和束状带细胞质 ,肾中HSP70阳性免疫反应肾小管较肾小球强 ,肾髓质较肾皮质强。 结论 热休克反应中不同组织器官HSP70的表达存在明显差异性 ;HSP70合成迅速且降解缓慢 ;
Objective To study the expression of HSP70 in the mouse cerebrum,pituitary gland,kidney,adrenal gland during the recovery period. Methods 176 Kunming mice were divided into control group and experiment groups at random.The experiment group:1.Subjected to heat shock under 40℃,42℃,44℃,46℃ for 30*!min respectively,then recovered for 2*!h and 8*!h;2. Subjected to heat shock under 46℃ for 30*!min,recovered for 2*!h,4*!h,6*!h,8*!h,12*!h,24*!h,48*!h,72*!h,the expression of HSP70 was observed with SP immunohistochemical method and image analysis technique. Results 1.Heat shock could induce the experession of HSP70 in kidney and adrenal at 42℃,44℃,46℃,induce the expression of HSP70 in cerebrum and pituitary gland at 44℃,46℃,and the maximum areas of the positive immunoreactivity(IR)in both were under 46℃(P<0 01);2.The synthesis of HSP70 reached peak 4-8*!h after heat shock in cerebrum and pituitary gland (P<0 01),8-12*!h after heat shock in kidney and adrenal gland(P<0 01);3.The positive IR of HSP70 was localized in neurolemma and nodes of Ranvier in cerebrum,in neurolemma in hypophysis pars nervosa,in cytoplasm of the renal tubule,adrenal reticularis and fasciculate zona.In kidney,the positive HSP70-IR was stronger in the renal tubule cells than that in the glomerular cells,stronger in medulla than that in the cortex.Conclusion The expression of HSP70 in different tissues and organs may be difference in heat shock response.The synthesis of HSP70 may be rapid and its degradation may be slow;The cerebrum and pituitary gland may have a stronger heat shock capacity.
