摘要
对黄精中总黄酮的含量进行测定和性质分析。用超声波法提取黄精中的总黄酮,高效液相色谱法(HPLC法)测定黄精总黄酮的含量,检测不同条件下黄精总黄酮在270 nm处的吸光度(A)的变化,研究其对pH、温度、光照的稳定性,采用ABTS法和FRAP法测定黄精总黄酮的抗氧化活性。黄精中含有黄酮,其含量为0.672 mg/g,在酸性到弱碱性环境(pH在2~8之间)中稳定性好,在温度低于80℃时具有好的热稳定性,ABTS法测得黄精黄酮抗氧化能力的半抑制率(EC50)为12.4μg/mL,FRAP法测定黄精总黄酮的相对抗氧化活性为2.27μg/mL。黄精中含有黄酮,且具有良好的光、热、酸碱稳定性和抗氧化活性。
Determine the content and analysis the properties of the total flavonoids of Rhizoma polygonati. Extract the total flavonoids from Rhizoma polygonati with ultrasonic method; determine the content of the total flavonoids of Rhizoma polygonati by ultraviolet spectrophotometry and HPLC. Its stability to pH, temperature, and light were studied in all different conditions by testing the absorbance in 280 nm. It's anti-oxidation activity were studied by using ABTS and FRAP method. Rhizoma Polygonat contains flavonoids, whose content is 3.54 mg/g, the flavonoids of Rhizoma polygonati will be stable in the 2~8 range of pH and less than 80 ℃of temperature, EC50 is 12.4 μg/mL by using ABTS to test it's anti-oxidation activity, relative anti-oxidation activity value is 2.27 μg/mL by using FRAP method. Rhizoma polygonati contains flavonoids. The flavonoids of Rhizoma Polygonat has good stability to the light, heat and acid-base, it has good antioxidant activity.
