摘要
目的:分析冷饭藤挥发油的化学成分并探讨对人乳腺癌MCF-7细胞增殖的影响。方法:采用水蒸气蒸馏法提取挥发油,用气相色谱-质谱联用法(GC-MS)联合NIST08和WILEY275谱库检索,同时结合Kováts保留指数(Retention index,RI)对其化学成分进行定性。采用MTT法测定冷饭藤挥发油对人乳腺癌细胞MCF-7生长的抑制作用。结果:从冷饭藤挥发油中共鉴定出50个化合物,占所提取物挥发油总量的98.421%。挥发油对MCF-7细胞生长有抑制作用,且其作用具有浓度依赖性,半数抑制浓度(IC50)为377.61 mg·L-1。结论:冷饭藤挥发油主要成分为莰烯(17.770%)、龙脑(12.080%)、(1,7,7-三甲基降冰片烷-2-YL)乙酸(8.239%)和d-杜松烯(7.315%)。冷饭藤挥发油对人乳腺癌MCF-7细胞增殖起到抑制作用,且呈浓度依赖性。
Objective:To study the chemical components of essential oils from Kadsurae Oblongifoliae Radix and investigate their effect on proliferation of breast cancer cell line MCF-7.Method:The essential oil was extracted by steam distillation,and analyzed by GC-MS.Comparing with the data base NIST08 and WILEY275,and combing with Kováts retention index to identify the essential oil constituents.MTT assays were used to measure the inhibitory effect on the human breast cancer cell MCF-7.Result:Fifty compounds,account for 98.421% of the essential oils,were identified.The volatile oil has inhibitory effect on the growth of MCF-7 cells,and the effect is concentration dependant,IC50 377.61 mg ·L^-1.Conclusion:The main chemical constituents of the essential oils from Kadsurae Oblongifoliae Radix are camphene (17.770%),borneol (12.080%),bicyclo [2.2.1] heptan-2-ol,1,7,7-trimethyl-2-acetate (8.239%) and (+)-delta-cadinene (7.315%).And the essential oils of Kadsurae Oblongifoliae Radix can inhibit proliferation of MCF-7 cells,which shows concentration dependence.
