槐杞黄颗粒对过敏性紫癜肾炎大鼠蛋白尿及肾组织TGF-β1表达的影响 被引量:21

摘要 目的观察槐杞黄颗粒(槐耳菌、枸杞子、黄精)对过敏性紫癜肾炎大鼠血尿、蛋白尿和肾组织TGF-β1表达的影响,探讨其益气养阴的治疗作用及机理。方法将18只雄性SD大鼠用随机数字表法均分为空白组、模型组和槐杞黄组。模型组及槐杞黄组建立过敏性紫癜肾炎模型,槐杞黄组给予槐杞黄颗粒治疗。取大鼠血、尿及肾组织标本,用尿沉渣计数法检测尿红细胞个数;蛋白浓度测定试剂盒法(BCA)进行24 h尿蛋白定量;光镜下观察肾小球系膜变化;酶联免疫吸附法(ELISA)检测肾小球中TGF-β1的水平;聚合酶链反应(PCR)检测肾皮质TGF-β1 mRNA的表达。结果模型组镜下尿红细胞计数、24 h尿蛋白定量均显著高于空白组(前者P<0.05,后者P<0.01);与模型组比,槐杞黄组蛋白尿程度改善(P<0.05),尿红细胞计数虽有下降趋势,但差异无统计学意义,且大鼠肾组织病的系膜增生及炎性浸润较轻。ELISA结果显示槐杞黄组大鼠肾组织TGF-β1的水平降低,PCR表明TGF-β1 mRNA的表达升高,差异均有统计学意义(P<0.05)。结论槐杞黄颗粒的益气养阴作用能影响肾组织TGF-β1 mRNA的表达来降低过敏性紫癜肾炎大鼠血尿及蛋白尿水平,进一步改善肾脏炎症反应。 AIM To explore the effect of Huaiqihuang Granules ( Trametes robiniphila Murr, Lycii Fructus, Polygonati Rhizoma) on hematuria, proteinuria and renal tissue expression of TGF-beta 1 in rats with allergic pur- pura nephritis (Henoch - Schonlein purpura nephritis, HSPN) and discuss their therapeutic effect and the mecha- nism of tonifying qi and yin. METHODS A total of 18 SD male rats were divided into three groups randomly: blank group ( n = 6), model group ( n = 6) and Huaiqihuang Granules group ( n = 6). The latter two groups were built into IgA nephropathy model to simulate HSPN by the combination of BSA, LPS and CC14. After the molding and treatment, samples were taken from the blood, urine and nephridial tissue of rats. Red blood cells number in urine was tested by urine sediment counting method. Urinary protein ratio was measured by BCA method. The glo- merular mesangial changes were observed under light microscope. The expression of TGF-beta 1 in glomerulus was detected by ELISA method and the expression of TGF-beta 1 mRNA of renal cortical was detected by the PCR meth- od. RESULTS The microscopic red blood cells count and 24 h urine protein quantity in the model group wereobviously more than those in the normal group ( the former was P 〈 0. 05, the latter was P 〈 0. 01 ). Compared with the normal group, 24 h urine protein quantity significantly reduced ( P 〈 0.05 ) and red blood ceils count was less in Huaiqihuang group, but difference was not statistically significant. Mesangial proliferative changes and inflamma- tory infiltration of kidney tissues of rats in Huaiqihuang group reduced. ELISA results showed the TGF-beta 1 con- tent of Huaiqihuang group was lower as compared with the model group while TGF-beta 1 mRNA expression in Hua- iqihuang group increased (P 〈 0.05 ) by PCR. CONCLUSION Huaiqihuang Granules can tonify qi and yin and up-regulate the expression and distribution of mRNA of TGF-beta 1. It plays a key role in repairing tissues, de- creasing urinary protein and urine red blood cells count, and improving kidney inflammation.
