摘要
目的探讨miR-421在胃癌细胞中的表达水平及对BGC-823胃癌细胞增殖的作用。方法利用PCR方法分析miR-421在胃癌组织细胞及多种胃癌细胞株中的表达水平,并通过RNA转染技术将miR-421 mimics与miR-421 inhibitors转染BGC-823胃癌细胞;通过CKK-8试剂盒检测转染后BGC-823细胞的增殖情况。结果 miR-421在胃癌组织细胞及多种胃癌细胞株中均表达增强(P<0.05);miR-421 mimics转染BGC-823后,癌细胞增殖能力增强(P<0.05),而miR-421 inhibitors转染的BGC-823细胞则增殖力受到明显抑制(P<0.05)。结论 miR-421在胃癌组织细胞及多种胃癌细胞株中均过表达,而miR-421可能直接或间接参与调控BGC-823胃癌细胞的增殖。
Objective To investigate the expression of miR-421 in gastric cancer and its effect on cancer cell proliferation. Methods The expression level of miR-421 in gastric cancer tissues and gastric carcinoma cell lines were analyzed by PCR. miR-421 mimics or miR-421 inhibitors were transfected into gastric carcinoma BGC-823 cells. Cell proliferation in transfected BGC-823 cells was detected by CKK-8 kit. Results miR-421 was over-expressed in human gastric cancer cells and several gastric carcinoma cell lines( P 0. 05). The growth of BGC-823 cells was promoted significantly after transfected with miR-421 mimics( P〈 0. 05),while the proliferation was inhibited after transfected with miR-421 inhibitors( P〈 0. 05). Conclusion miR-421 is over-expressed in gastric carcinoma cells and miR-421 may promote the proliferation of gastric carcinoma BGC-823 cells.
