Notch信号分子在ICA诱导小鼠胚胎干细胞向心肌细胞分化的作用 被引量:4

摘要 目的观察Notch信号通路在淫羊藿苷(ICA)诱导胚胎干细胞(ESC)向心肌细胞分化过程中的作用。方法实验分为自然分化组、ICA诱导分化组及ICA+MW167(Notch信号抑制剂)组。相差显微镜下观察各组自发性节律收缩细胞出现情况的差别,免疫荧光染色观察各组细胞心肌特异性蛋白ANP的表达,RT-PCR检测各组细胞ANP mRNA的表达。结果 ICA+MW167组自发性节律收缩细胞数明显多于自然分化组与ICA诱导分化组,ICA+MW167组心肌细胞特异性蛋白ANPmRNA和蛋白的表达水平随培养时间延长逐渐增加,明显高于自然分化组与ICA诱导分化组。结论 Notch信号通路参与了ICA诱导ESC分化为心肌细胞的过程,抑制该信号通路可以明显提高心肌细胞特异性ANP基因及蛋白的表达,上调分化的心肌细胞比率。 Objective To study the action of Notch signaling molecule in differentiation of murine embryonic stem cells(ESCs) into cardiomyocyte induced 3y icarrin. Methods The murine ESCs(ES-EI4) were divided into three groups: control, ICA group and ICA+MW167(Notch inhibition). The expression of ANP protein and mRNA in ESCs was determined by immunofluorescenee staining and RT-PCR respectively. Results The number of the beating ESCs was significantly increased in ICA+MW167 group than in ICA group and in control group. The expression levels of ANP protein and mRNA in ESCs were higher in ICA+MW167 group than in ICA group and in control group. Conclusion MW167 can promote the differentiation of embryonic stem ceils to cardiomyoeytes.
