白藜芦醇通过抑制纤溶亢进延长大鼠异种心脏移植物的生存期

Resveratrol inhibits hyperfibrinolysis and prolongs heart xenograft survival time in hamster-to-rat model

摘要 目的 观察白藜芦醇(RES)对仓鼠→大鼠异种心脏移植发生的延迟性异种排斥反应(DXR)中血液高凝状态的影响.方法 建立仓鼠→大鼠异种心脏移植模型,采用来氟米特(LEF)和FK506联合用药14 d建立DXR模型,并使用RES进行后续治疗.观察移植心脏的存活时间、病理以及凝血6项.结果 对照(Ctrl)组平均生存期为8.5d,而RES组平均生存期达到23.5 d.RES组有效地减少血管壁周围炎性细胞浸润以及血管内血栓的形成,并有效地降低纤维蛋白原(FIB-c)以及纤溶降解产物D-二聚体(D-D)水平,改善血液的高凝状态.结论 RES能通过降低FIB-c的产生以及降低纤维蛋白降解降解产物D-D的水平,改善血液的高凝状态,延长移植物的生存期. Objective To study the inhibitory effects of resveratrol (RES) on hyperfibrinolysis in delayed xenograft rejection (DXR).Methods Hamster hearts were transplanted into Lewis rats receiving an accommodation-inducing treatment containing leflunomide and FK506.Then all of the rats were divided into two groups randomly.The control group received vehicle (1% carboxymethyl cellulose),and the treated groups received RES 100 mg/kg,by daily gastric gavage until rejection.The survival of transplants was recorded.We also observed the histological sections of the graft and the coagulation factor of the recipents.Results The meam survival time (MST) in RES group was 23.5 d and 8.5 d in control group.The intravascular thrombosis,interstitial hemorrhage,and polymorphonuclear cell infiltrtion were found in control group.In contrast,the RES group showed normal vessels without endometrial hyperplasia and reduced immune cell infiltration,and maintained a healthy myocardium.The level of fibrinogen (FIB-c) and D-D in the RES group was reduced as compared with control group.Conclusion RES prolongs xenograft rejection survival after withdrawal of LEF and FK506 by improving blood hypercoagulable state,and reduing the level of FIB-c and D-D.
