摘要 目的:本文旨在观察没食子儿茶素没食子酸酯(EGCG)对中波紫外线(UVB)引起的表皮HaCaT细胞损伤,以及对细胞着色性干皮病蛋白A(XPA)和切除修复互补交叉蛋白1(ERCC1)蛋白表达的影响。方法:以30mJ/cm2 UVB照射HaCaT细胞,照光前后加入50g/mL EGCG进行干预,Western blot法检测细胞XPA和ERCC1蛋白的表达水平。结果:UVB照射后细胞内XPA和ERCC1蛋白表达水平逐渐增高,在照光后4h达到峰值,后逐渐恢复至接近正常值;加入EGCG干预可明显降低XPA和ERCC1蛋白的表达水平。结论:EGCG对紫外线光损伤的干预作用可能与调控细胞内XPA和ERCC1蛋白的表达有关。 Objective To investigate the effects of EGCG on photodamage induced by UVB irradiation XPA and ERCC1 proteins expression in HaCaT cells. Methods HaCaT cells were irradiated with 30mJ/cm2 UVB.50 g/mL EGCG was added into the medium before or after UVB irradiation. Western blot assay was used to measure XPA and ERCC1 expression. Results 30mJ/cm2 UVB irriadiation increased the XPA and ERCClexpression which arrived at the peak at 4h after UVB irradiation.EGCG treatment significantly inhibited the XPA and ERCClexpression. Conclusion The effects of EGCG on UVB induced the photodamage in HaCaT cells maight be related with the inhibitory efficency of EGCG on XPA and ERCC1 expression.
