慢性低灌注大鼠基质金属蛋白酶和淀粉样物质的表达

Expression of Matrix Metalloproteinases-2,9 and β-amyloid in Rats after Chronic Brain Hypoperfusion

摘要 目的:研究慢性低灌注后基质金属蛋白酶MMP-2、MMP-9和淀粉样物质Aβ40表达的变化。方法:Wistar大鼠48只随机分为假手术组和缺血组各24只,缺血组大鼠结扎双侧颈总动脉,假手术组不造成缺血。免疫组化染色观察MMP-2、MMP-9和Aβ40表达的变化。结果:缺血组大鼠低灌注后7 d,免疫组化染色显示MMP-2和Aβ40在皮质的静脉内皮上有阳性表达;低灌注后14 d,MMP-2和Aβ40在较小动脉内皮上阳性表达;低灌注后30 d在相对较大的动脉内皮上阳性表达;MMP-9在血管内皮未见表达。结论:慢性低灌注后Aβ40在血管内皮沉积,MMP-2表达增加。 Objective:To study the expression of Matrix metalloproteinases (MMP-2, MMP-9) and β-amyloid (Aβ40) in the brain of rats with chronic hypoperfusion. Methods:Fourty-eight Wistar rats were randomly divided into sham group and ischemic group (24 rats in each group). For the rats in the ischemic group, the bilateral common carotid arteries were ligated. The rats in the sham group received similar operation without occlusion in the the bilateral common carotid arteries. Immunohistochemical staining was performed to investigate the expres-sion of MMP-2, MMP-9 and Aβ40. Results: In the ischemic group, the expression of MMP-2 and Aβ40 was observed in vein endothelium at 7 days after chronic hypoperfusion, in small artery endothelium at 14 days after chronic hypoperfusion, and in large artery endothelium at 30 days after chronic hypoperfusion. The expression of MMP-9 in ischemic group was not observed in blood vessel endothelium. Conclusion:Chronic brain hypoperfu-sion in rats can result in the deposition of Aβ, and the alteration of the expression of MMP-2.
