摘要
目的探讨血管易感基因瘦素、瘦素受体以及信号转导和转录活化因子3(STAT3)在脑动静脉畸形(AVM)破裂出血的表达变化及其影响。方法选择脑AVM病人80例,随机分为出血组(n=40)和未出血组(n=40)。应用实时定量PCR、Western印迹法检测瘦素、瘦素受体以及STAT3在脑AVM中的表达。结果实时定量PCR与Western印迹法检测监测结果显示:瘦素、瘦素受体以及STAT3在出血组和未出血组间的表达,差异均具有显著统计学意义(均P<0.01)。结论在分子生物学水平上,研究瘦素、瘦素受体以及STAT3对脑AVM破裂出血的影响,对于预测和减少脑AVM破裂出血的风险和临床诊疗具有重要作用。
Objective To investigate the changes in expressions of vascular susceptibility gene leptin, leptin receptor and signal transduction and transcriptional activation factor 3 (STAT3) in intracerebral hemorrhage from arteriovenous malformation (AVM) rupture and their effects. Methods Eighty patients with cerebral AVM were randomly divided into bleeding group (n = 40) and nonbleeding group (n = 40). The real-time quantitative PCR and Western blotting technique were used to detect leptin, leptin receptor and STAT3 expressions in cerebral AVM. Results The results from real-time quantitative PCR and Western blotting showed that there were significant differences in leptin, leptin receptor and STAT3 expressions between bleeding and non-bleeding groups, with a significant statistical significance (all P 〈0.01). Conclusions Studies at a level of molecular biology about the influences of leptin and leptin receptor and STAT3 on intracerebral hemorrhage due to AVM rupture play an important role in predicting and reducing the risk of intracerebral hemorrhage due to AVM rupture and in clinical diagnosis and treatment of the hemorrhage.
