摘要
目的:呼吸代谢是能量支出的一部分,也是动物营养物质利用与分配的基础,若个体能量支出增加,则可能导致寿命减少。本研究通过测定不同浓度决明子水煎液对家蚕呼吸代谢的影响,探明决明子抗衰老作用的机制。方法:实验时,取龄期相同、生长整齐的三眠蚕随机分为5组,即空白对照组,决明子水煎液0.65 g/ml(生药浓度)剂量组、0.35 g/ml剂量组、0.15 g/ml剂量组,维生素E(VE)0.0015 g/ml剂量组(阳性对照),每组30条。使用涂抹法将不同浓度药液涂抹于新鲜桑叶上,4龄期内喂食3次,5龄期内喂食6次,在五龄期末测定动物呼吸代谢及体重。结果:与对照组及维生素E对照组相比,各给药组家蚕体重有显著降低。同样,家蚕单位体重代谢率及整体代谢率在各给药组均有不同程度降低,其中决明子0.65 g/ml剂量组达到了极显著水平,分别为RMR(ml O2/g·h)=1.37±0.34,PC=0.001,RMR(ml O2/只·h)=1.86±0.46,PPC=0.004。皮尔逊相关分析结果表明,整体代谢率与体重呈现显著正相关关系r=0.275,P=0.001。单位体重代谢率与体重的相关关系不明显r=-0.030。整体代谢率(r=-0.392,P<0.001)和单位体重代谢率(r=-0.325,P<0.001)与家蚕寿命之间的关系均呈现显著负相关。结论:决明子水煎液喂食后家蚕代谢率降低,并导致寿命增加。
Rest metabolic rate (RMR) is part of energy expenditure, and also is a character of animalg life. Moreover, it provides the base of en- ergy allocation for animal. High metabolic rate may provide a performance advantage, but it also entails a physiological cost. A long-held as- sumption is that high mass-specific energy expenditure is associated with short lifespan. To examine the relationship between energy expendi- ture and life span, this paper explore the anti-aging effect of different concentration of Catsia tora L. water extract on rest metabolic rate of Bombyx mori L. to test this hypothesis. During experiment, the 150 Bombyx moil L. with the same age and growth were divided into five groups included that control group, High dose (0.65 g/ml), middle dose (0.35 g/ml), low dose (0.15g/ml) and positive group (VE). Physic liquor was daubed on the mulberry leaf,3 times during 4th instars and 6 times during 5th instars. At last in 5 instars, RMR was meas- ured by the CYES-H O2-CO2 instrument, body mass was also measured at the same time. The results show that body mass in each group de- crease significantly (P 〈0.01, Turkey HSD) compared with control group and positive group (VE). RMR per gram (ml O2/g - h) and RMR (ml O2/h) also decreased in each experiment group compared with control group and positive group (VE), RMR( ml O2/g - h) = 1. 37 + 0.34, PC = 0,001, RMR ( ml O2/ · h) = 1.86 + 0.46, PPC = 0. 004 respectively. There was significant positive relationship between RMR(ml O2/ · h) and body mass (Pearson correlation, r=0. 275, P = 0. 001 ), there was no significant relationship between per gram RMR(ml O2/g · h) and body mass (r = -0. 030). However, there were significant relationships between RMR (ml O2 · h), per gram RMR ( ml O2/g · h) and life span of Bombyx moil L. the correlation coefficient was rRMR ( ml O2/· h) = -0. 392, P 〈 0.001 and r RMR ( ml O2/g · h) = 4). 325, P 〈0.001 respectively. So, it concludes that the effect of Catsia tora L water extract can decrease the oxygen con- sumption and increase the life span of Bombyx moil L. These conclusions support the anti-aging of Catsia tora L and need to further research.
