保肾颗粒对庆大霉素所致大鼠急性肾功能衰竭的保护作用 被引量:12

摘要 目的:观察复方保肾颗粒对庆大霉素(GM)致大鼠急性肾功能衰竭(ARF)的保护作用并初步探索其可能存在的部分作用机制。方法:Wistar雄性大鼠96只随机分为空白对照组、模型对照组、肾炎康复片(阳性药)组和复方保肾颗粒3.6g/kg、1.8 g/kg、0.9 g/kg剂量组,每组16只。采用腹腔注射庆大霉素(GM)法制备急性肾功能衰竭模型,各组在造模期间同时给予相应药物,连续给药14天。实验第6天和第13天收集大鼠24h尿液,记录尿量并检测尿N-乙酰-β-氨基葡萄糖苷酶(NAG酶);分别在给药第7天和第14天末次给药后4h分别处死8只动物,检测各组血肌酐(Scr)、尿素氮(BUN);取肾脏称重,计算肾脏系数,测肾皮质匀浆MDA含量和SOD、CAT酶活性并作HE染色观察肾脏病理改变。结果:与模型对照组相比,复方保肾颗粒3.6g/kg、1.8 g/kg剂量组能明显减少尿量,降低尿NAG活性,减少血Scr及BUN的含量,明显降低MDA水平,提高SOD和CAT酶的活性,降低肾脏系数;组织病理学检查结果显示复方保肾颗粒3.6g/kg、1.8 g/kg剂量组肾小管上皮细胞染色、形态明显好于模型对照组。结果还表明随着给药时间的延长,复方保肾颗粒对急性肾功能衰竭的改善保护作用愈加显著。结论:复方保肾颗粒对庆大霉素引起的急性肾功能衰竭具有一定的保护作用;其作用机理可能与其减轻肾损伤、抗氧化及促进修复再生等作用相关。 To investigate the protective effect and the mechanism of Fufangbaoshen Granule on gentamicin-indueed acute renal failure (ARF) in rats. Methods: 96 healthy male Wister rats were randomly divided into 6 groups, 16 rats per group: the normal group, the model group, Shenyankangfu Trache group, and Fufangbaoshen Granule 3.6g/kg, 1.8g/kg and 0.9g/kg groups. The acute renal failure model was developed by intraperitoneal injection of gentamicin (GM). After 14 days gavage administration, 8 rats were executed respectively on the 7th day and the 14th day. The changes of serum ereatinine (Scr) , blood urea nitrogen (BUN) , urinary N-acetyl-J3-amino glyeosidase enzyme ( NAG), total urinary volume in 24h, and oxidative stress-related indexes such as SOD, MDA and CAT were detected. The renal indexes were calculated and the changes of renal histopethological were observed too. Results: Compared with model group, Fufangbaoshen Granule 3.6g/kg group and I. 8g/kg group can significantly reduce the urine volume and the activity of NAG, as well as the contents of Ser and BUN, obviously improves the activity of SOD and CAT, lower the level of MDA and kidney indexes. The results of histopathological observation show renal tubular epithelial cells of Fufangbaoshen Granule 3.6g/kg group and I. 8g/kg group are obviously better than those of the model group. The results also show that the longer the period of garage administration is, the more remarkable the improvement of Fufanghaoshen Granule for ARF will be. Conclusions: Fufangbaoshcn Granule have certain protection for ARF caused by GM ; the mechanism may he relat- ed with those functions, such as relieving renal hurt, antioxidation, and promoting restoration and regeneration.
