运动点电针对急性脊髓损伤大鼠半胱氨酸天冬氨酸蛋白酶3和降钙素基因相关肽表达的影响 被引量:10

摘要 目的:探讨运动点电针对急性脊髓损伤大鼠半胱氨酸天冬氨酸蛋白酶3(caspase-3)和降钙素基因相关肽(CGRP)表达的影响。方法:将60只SD大鼠随机分为4组,每组15只。A组:运动点电针组;B组:非运动点电针组;C组:模型组;D组:假手术组。采用改良的Allen法建立大鼠急性脊髓损伤模型。A、B组分别于术后立即开始下肢运动点和非运动点进行电针治疗;C、D组不做任何治疗。各组大鼠分别于1、4、7d行BBB评分,通过免疫组织化学方法(IHC)观察各组caspase-3和CGRP阳性细胞的表达情况。结果:60只SD大鼠均进入结果分析。BBB评分结果显示:A组评分明显高于B、C组(P<0.05);免疫组化结果显示:1caspase-3的表达:损伤后各组均可见阳性细胞表达,D组仅见少量的阳性细胞。其中A、B组阳性细胞表达明显低于C组(P<0.05);而A组的表达又明显低于B组(P<0.05);2CGRP的表达:各组均可见阳性细胞表达,A、B组明显高于C组(P<0.05),而A组又明显高于B组(P<0.05)。结论:运动点电针可抑制caspase-3和促进CGRP的表达,可能有助于脊髓功能的恢复。 Objective: To discuss the impact of electroacupuncture at motor points on caspase-3 and calcitonin gene related peptide(CGRP) expressions of rats with acute spinal cord injury. Method: Sixty SD rats with spinal cord injury were randomly divided into four groups, 15 rats in each group:group A: electroacupuncture at motor points; group B: electroacupuncture at non-motor points; group C: model group; group D: sham-operated group. Modified Allen's method was used to establish rats model of acute spinal cord injury. Rats in groups A and B acupuncture therapy were administered at motor points and non-motor points immediately after spinal cord injury. Rats in groups C and D received no treatment. At day 1,4 and 7, BBB(Basso-Beattie-Bresnahan) scores were assessed in each group,Then the expressions of caspase-3 and CGRP positive cells of rats after acute spinal cord injury in each group were observed by immunohistochemistry method. Result: BBB scores:In group A BBB scores were significantly higher than that in groups B and C(P〈0.05). Immunohistochemical results:①Caspase-3: The expressions of positive cells emerged in all groups after injury, in group D only small amount of positive cells emerged.The expressions of positive cells in groups A and B were significantly lower than that in group C (P〈0.05),and the expression of positive cells in group A was lower than that in group B (P〈0.05).②CGRP:The expressions of positive cells emerged in all groups. The expressions of positive cells in groups A and B were significantly higher than that in group C(P〈0.05), and the expression of positive cells in group A was higher than that in group B (P〈0.05). Conclusion: Electroacupuncture at motor points can suppress the expression of caspase-3 and promote the expression of CGRP and would be helpful for the spinal cord function recovery.
