复方茵丹汤对大鼠急性肝内胆汁淤积的干预作用 被引量:12

The effect of compound Yindan decoction in rat model with acute intrahepatic cholestasis

摘要 [目的]观察复方茵丹方对急性肝内胆汁淤积(IC)大鼠模型的治疗作用及其作用机制。[方法]选择雄性wistar大鼠45只,随机分为正常组、模型组、中药组3组。应用α-萘异硫氰酸酯(ANIT)灌胃诱导大鼠IC模型。中药组于造模同时给予复方茵丹方灌胃。分别于造模后24h、48h、72h测量大鼠体重、肝重,采集大鼠血清、肝组织标本。[结果]与同期模型组比较,中药组大鼠活动量、进食及尿色均有明显改善,且中药组大鼠的体重增高,肝重、肝指数降低,于72h时差异有统计学意义(P<0.05);肝功能结果显示,中药组在72h各指标明显降低(P<0.05);肝脏病理结果显示,中药组肝组织于72h中性粒细胞浸润明显减轻。[结论]复方茵丹方能够改善肝内胆汁淤积大鼠的一般状况,降低血清胆红素水平、降低转氨酶、胆管酶,保护肝脏功能,减轻肝细胞肿胀和毛细胆管炎症,改善肝脏组织学病变,对ANIT诱导的IC具有一定疗效。 [Objective]To investigate the treatment effect and mechanism of compound Yindan decoctionin rat model with acute intrahepatic cholestasis (IC). I-Methods]Forty-five male Wistar rats were randomlydivided into three groups. Rat models of acute intrahepatic cholestasis were induced by alpha-naphthyl iso-thioeyanate(ANIT). The rats of TCM group were given compound Yindan decoction by gavage at modelingtime. Weight,liver weight,serum and liver tissue of rats were collected at modeling 24 h,48 h,and 72 h.[Results]Compared with model group,the activity,food intake and urine color of rats in TCM group weresignificantly improved. Meanwhile, body weight was increased and liver weight and liver index were de-creased. The difference was statistically significant in 72 h(P〈0.05). All the indexes of liver functionwere decreased significantly in the TCM group at 72 h (P〈0.05). Liver pathology showed that neutro-phils infiltration of liver tissue was significantly alleviated in TCM group at 7Z h. [Conclusion]The compoundYindan decoction can improve the general situation of rats with intrahepatic cholestasis, reduce the levels of serumbilirubin, bile duct and transaminase enzyme, protect the liver function, reduce the swelling of the liver cells and bilecapillary pathological changes of liver tissue inflammation,indicating a certain effect on IC induced by ANIT.
