摘要
建立一种高效液相色谱-蒸发光散射检测(HPLC-ESLD)同时测定葡萄干中的果糖、葡萄糖、蔗糖和麦芽糖的方法。采用氨基柱进行分离,乙腈-水为流动相,流速为1.0 mL/min,ELSD漂移管温度为85℃,辅气为氮气,雾化气体压力为40 psi。结果表明,在该条件下,4种糖的组分都能够较好地分离,线性良好,该方法相对标准偏差(RSD)1.2%-2.1%,平均回收率98.25%-100.7%,4种组分的线性相关系数r〉0.9990。测定了新疆8种不同品种葡萄干中果糖、葡萄糖、蔗糖和麦芽糖的含量。
A method(HPLC-ELSD) for simultaneous measurement of the content of fructose/glucose/sucrose/maltose in raisin had been developed. The carbohydrates were separated on NH2 column by using acetonitrile-water as the mobile phase with the flow rate as 1 mL/min and the drift tube temperature at 85 ℃,nitrogen was used as auxiliary gas and the pressure of atomization gas was 40 psi. As a result, the above four sugar components in raisin were separated successfully and the linearity was good. The relative standard deviation was 1.2 %-2.1 %, the average recoveries were 98.25 %-100.7 % and the linear correlation was r〉 0.9990. In the experiments, the content of frucotse/glucose/sucrose/maltose in 8 different species of raisin in Xinjiang was measured.
