响应面分析法优化黄芩总黄酮提取工艺的研究 被引量:2

Study on Optimization of the Extraction Techniques of the Total Flavonoids from Scutellariae baicalensis Using Response Surface Methodology

摘要 目的优选超声波法提取黄芩总黄酮的工艺条件,为工业化生产黄芩总黄酮提供参考。方法采用超声波法提取黄芩总黄酮,对使用Al(NO3)3-NaNO2显色法测定黄芩总黄酮进行方法学考察;选取乙醇浓度、液固比、提取时间3个影响因素进行BoxBenhnken中心组合设计,利用响应面分析法(RSM)对提取工艺参数进行优化。结果方法学考察结果表明本实验室采用Al(NO3)3-NaNO2显色法测定黄芩总黄酮及超声提取工艺可靠;提取黄芩总黄酮最佳提取工艺为:乙醇浓度60%,液固比40mL/g,提取时间45min;预测值为1.776%,实际得率为1.704%,两者吻合度较高。结论 Box-Benhnken设计结合响应面分析法对黄芩总黄酮提取工艺进行优化既方便又快捷,得出的参数可信度高、实用性好。 Objective Optimizing the ultrasonic extraction techniques of the total flavonoids from Scutellariae baicalensis,to provide a reference for industrial total flavonoids extraction of Scutellariae baicalensis. MethodUsing the ultrasonic method to extract total flavonoids in Scutellariae baicalensis; investigating the methodology of total flavonoids Scutellariae baicalensis measured by Al( NO3)3- NaNO2reaction; ethanol concentration,liquid- solid ratio and extracting time selected in Box- Benhnken central composite design; optimizing extraction process parameters by using the response surface method( RSM). Result The methodological evaluation showed that it was reliable to measure the total flavonoids by Al( NO3)3- NaNO2 reaction and the ultrasonic extraction technology in this laboratory; The optimum technical condition in extracting total flavonoids of Scutellariae baicalensis; ethanol concentration 60%,liquid- solid ratio 40 mL /g,extraction time 45 minutes. The predictive value was 1. 776% and the yield of the total flavonoids was actually 1. 704%,and both results were in good agreement. Conclusion Box- Benhnken design combined with response surface method was convenient and reliable for extracting total flavonoids from Scutellariae baicalensis. The optimal extraction parameters was of reliability and practicality.
