摘要
目的探讨大承气汤对不完全性肠梗阻大鼠白介素-1(IL-1)和白介素-6(IL-6)及回盲部形态学的影响。方法将60只SD大鼠随机分为空白对照组,模型对照组,假手术组,大承气汤高、中、低剂量组(以下简称高、中、低剂量组)。除空白对照组外,假手术组造成手术创伤,其余大鼠制备成不完全性肠梗阻模型,空白对照组、模型对照组、假手术组给予相同剂量的生理盐水灌胃,高、中、低剂量组分别以1.16、0.58、0.29 g/mL灌胃治疗。(1)运用酶联免疫法(ELISA)检测大鼠血清中IL-1、IL-6含量;(2)对大鼠回盲部进行HE染色并观察其形态学变化。结果(1)与模型对照组比较,高、中剂量组血清中IL-1、IL-6含量明显降低(P<0.01);(2)高、中、低剂量组与模型对照组大鼠回盲部黏膜损伤评分差异有统计学意义(P<0.01),高剂量组与中、低剂量组大鼠损伤评分比较,差异有统计学意义(P<0.01)。结论大承气汤降低不完全性肠梗阻血清IL-1以及IL-6含量,保护不完全性肠梗阻肠道上皮细胞,具有肠屏障保护功能。
Objective To explore the effects of Dachengqi Decoction on serum interleukin-1(IL-1), interleukin-6(IL-6) and ileocecal morphologyin in incomplete intestinal obstruction rats models. Methods 60 SD rats were randomly divided into blank control group, model control group, sham-operated group, high, medium and low dose Dachengqi decoction group. In addition to the normal control group, the sham-operated group needle penetration mesenteric without ligation, the other groups were caused by incomplete intestinal obstruction models. The blank control group, model control group, sham-operated group were given the same dose of normal saline. The high, medium, low dose groups were respectively treated by gavage 1.16,0.58, 0.29 g/mL.(1) Using enzyme linked immunosorbent assay(ELISA) detected the content of Interleukin-1,(IL-1), interleukin-6(IL-6) of serum in rats.(2) The morphological changes of cecum in rats were observed with. HE-staining. Results(1) Compared with the model control group, the high and medium dose groups of IL 1 and IL-6 in serum were significantly lower, there was statistically significant(P〈0.01).(2) Compared with the model group,the high, medium and low dose groups rats of ileocecal mucosal injury score, the differences were statistically significant(P〈0.01). Compared with the high dose group, the medium and low dose group of rats in ileocecal mucosal injury score, the differences were statistically significant(P〈0.01). Conclusion The model control group of IL-1 and IL-6 levels in rats serum increased. Dachengqi Decoction reduced serum IL-1 and IL-6 content in incomplete intestinal obstruction rats. Dachengqi decoction can reduce IL-1 and IL-6 content and protect the intestinal epithelia cells of incomplete intestinal obstruction in rats.
