摘要
目的:研究六神丸的体外抗肝癌活性作用及其有效部位。方法:用高效液相色谱法(HPLC)分析六神丸的不同溶媒提取物成分;四甲基偶氮唑蓝(MTT)比色法检测六神丸的不同溶媒提取物对人肝癌细胞HepG2和人正常肝细胞L02生存的影响。结果:六神丸各提取物均能抑制HepG2细胞生存,其中醇提上清液的抑制作用(IC50=14.8μg·ml-1)显著强于水提上清液(IC50=279.4μg·ml-1),显示出较强的体外抗肝癌活性效果;并且醇提上清液对L02细胞生存无明显抑制作用(IC50=111.1μg·ml-1)。采用HPLC分段收集醇提上清液,MTT检测结果显示,各分段收集液对HepG2细胞的抑制作用(IC50分别为2 315.5、68.3、1 033.3μg·ml-1)均弱于醇提上清液。结论:六神丸具有体外针对性地抑制癌细胞生存抗肝癌活性作用,其最佳有效部位为醇提上清液。
OBJECTIVE To investigate the anti-liver cancer activity of Liu-Shen-Wan in vitro. METHODS The effects of Liu-Shen-Wan and its effective fractions on the proliferation of human hepatoeellular carcinoma cells (HepG2) and normal hu- man liver cells (L02) were detected by a MTT assay. The component analysis was carried out using a high performance liquid chromatography (HPLC) technique. RESULTS The MTT assay results showed that all extracts of Liu-Shen-Wan inhibited the proliferation of HepG2 cells. The ethanol supernatant extract displayed the most significant effect with its half-maximal inhibi- tory concentration (IC40 = 14. 8μg·ml-1 ), in comparison to the water supernatant extract (IC50 = 279. 4 μg·ml-1 ). Moreover, the ethanol supernatant extract on the L02 cell proliferation had no obvious inhibitory effect (IC40 = 111.1 μg·ml- 1 ). The ethanol supernatant extract was further divided into three fractions by a preparative high-performance liquid chromatography. However, the inhibitory effect of three fractions on HepG2 cell inhibitory effects (IC50 = 2315. 5,68. 3 and 1 033. 3 μg· ml 1, re- spectively) were weaker than the ethanol supernatant extract. CONCLUSION Liu-Shen-Wan exhibited the activity of anti-liver cancer in vitro, and the best effective parts were the ethanol supernatant extract.
