摘要
目的:通过对转基因小鼠基因型的鉴定及配种繁育,得到一定数量带有目的基因的小鼠,建立转基因小鼠库。方法:利用PCR法对小鼠基因型进行鉴定,采用经典育种与PCR相结合的方法对转基因小鼠进行繁育。结果:Fmr1/PCPcre双转小鼠共产仔14窝,总计91只,阳性有70只,阳性率为76.92%;Fmr1/PV-cre双转小鼠共产仔21窝,总计164只,阳性有126只,阳性率为76.83%;chat-cre小鼠共产仔19窝,总计102只,阳性有40只,阳性率为39.22%;Ai35小鼠共产仔11窝,总计75只,阳性有29只,阳性率为38.67%;Ai14小鼠共产仔7窝,总计40只,阳性有26只,阳性率为65.00%。结论:成功繁育鉴定出实验所需数量的阳性转基因小鼠,并对转基因小鼠的鉴定及繁育建系方式进行了优化,建立了转基因小鼠库,为深入研究小鼠的生理性功能提供了良好的实验动物模型。
Objective: To establish a transgenic mouse library. Methods: To breed the transgenic mice by classical breeding methods and identify the mouse genotypes by PCR. Results:14 litters and 91 Fmrl/PCP-cre double transgenic mice were breeded, of which 70 mice were positive, the positive rate was 76.92% ; 21 litters and 164 Fmrl/PV-cre double transgenic mice were breeded, of which 126 mice were positive,the positive rate was 76.83% ; 19 litters and 102 chat-cre mice were breeded, of which 40 mice were positive, the positive rate was 39.22% ; 11 litters and 75 Ai35 mice were breeded, of which 29 were positive, the positive rate was 38.67% ;7 litters and 40 Ail4 mice were breeded, of which 26 mice were positive, the positive rate was 65.00%. Conclusion : The needed quantity of positive transgenic mice were successfully breeded and the transgenic mouse library which will be a good experimental animal model for the study of the physiological function of mice was established.
