摘要
目的:建立披针叶黄华不同部位中金雀花碱及其衍生物的高效液相色谱分析方法。方法:采用Lichrospher Si-120A色谱柱(250 mm×4.6 mm,5μm),以甲醇-乙醚-5%氨水(46∶52∶2)为流动相,流速1.0 mL·min-1,检测波长310 nm,柱温为室温。结果:金雀花碱与其2个衍生物完全分离,金雀花碱、N-甲基金雀花碱和N-甲酰基金雀花碱在各自的线性范围内线性关系均良好(r≥0.9999),最低检测限分别为0.5、0.1、0.3μg·mL-1,分别测定了披针叶黄华不同部位中3个成分的含量。结论:该方法可用于披针叶黄华中金雀花碱及其衍生物的测定,为金雀花碱生产原料的采收提供了依据。
Objective : To establish an HPLC method for determination of cytisine and its two derivatives in the dif- ferent parts of T. lanceolata R. Br.. Methods:The determination was performed on a Lichrospher Si - 120A column (250 mm × 4.6 mm,5 μm). The mobile phase was composed of methanol, diethyl ether and 5% ammonia solution (46: 52: 2)at the flow rate of 1.0 mL · min-1 ,the detection wavelength was 310 nm and the column temperature was room temperature. Results: Cytisine and its two derivatives were completely separated. The linearities of the three compounds were good in their own linear ranges ( r ≥ 0. 9999 ). The LOD of cytisine, N - methylcytisine, N - formylcytisine were 0.5,0. 1,0.3 μg · mL-1 respectively. The contents of three compounds in different parts of T. lanceolata were determined by the established method. Conclusion:This method can be used to determine the con- tents of cytisine and its two derivatives in the different parts of T. lanceolata. The results of the study provides refer- ence for procurement and selection of the raw material for eytisine.
