HPLC-ELSD及GC-MS法分析参麦注射液及中间体糖类成分 被引量:6

Analysis of saccharides of Shenmai injection and its intermediates by HPLC-ELSD and GC-MS method

摘要 目的:对参麦注射液及其中间体的糖类化合物进行定性分析。方法:首先采用HPLC-ELSD结合Alltech PrevailTMCarbohydrate ES色谱柱(250 mm×4.6 mm,4μm)对糖类型做初步判断,流动相为乙腈-水(70∶30),再利用Agilent气相色谱-质谱联用仪结合Agilent HP-5MS玻璃毛细管色谱柱(30 m×0.25 mm,膜厚0.25μm)对含有的糖类化合物进行分析。结果:HPLC-ELSD法可以鉴定出参麦注射液及其中间体仅含三糖以下的糖。GC-MS法进一步分析显示麦冬醇提液主要含葡萄糖、果糖、蔗糖3种糖,红参醇提液和参麦注射液中主要含葡萄糖、果糖、蔗糖、麦芽糖4种糖。结论:本法可以快速鉴定出参麦注射液及其中间体样品的糖类化学成分。 Objective: To conduct qualitative analysis of saccharides in Shenmai injection and its intermediates. Methods: Initial judgment was performed on the type of saccharides using HPLC - ELSD combined with Alhech Pre- vailTM Carbohydrate ES (250 mm× 4. 6 mm,4 μm) column with acetonitrile - water (70: 30) as the mobile phase ; and then saeeharides were analyzed using Agilent GC -MS combined with Agilent HP -5 MS (30 m × 0. 25 mm ,0. 25 μm) column. Results:The samples that were composed of saccharides which were less than trisaecharide can be accurately identified using HPLC - ELSD. The further analysis using GC - MS showed that the Ophiopogonis Radix intermediate contained glucose,fructose and sucrose ,and the Ginseng Radix et Rhizoma Rubra intermediate and the Shenmai injec- tion mainly contained 4 kinds of sugar compositions, i. e. , glucose, fructose, sucrose, and maltose. Conclusion: This method can quickly identify the carbohydrate chemical composition in Shenmai injection and its intermediate samples.
