摘要
目的:通过检测针刺后快速老化痴呆小鼠SAMP8海马脂筏蛋白质组的改变,阐述针刺治疗阿尔茨海默病的可能机理。方法:以SAMP8为研究对象,将其分为SAMP8组和针刺组,SAMR1为正常对照,DTT提取法提取海马脂筏,采用HPLC MS/MS shotgun analysis进行蛋白质鉴定,通过GO和KEGG分别对已鉴定蛋白质进行功能和信号分析。结果:SAMR1组鉴定出467种蛋白质,SAMP8组445种蛋白质,共有蛋白质为339种,SAMR1组独有蛋白质119种,SAMP8组106种。经针刺治疗后,SAMP8组独有的92种蛋白质经针刺后消失,103种蛋白质为针刺特异募集,其中36种同时出现于SAMR1组。对103种蛋白质进行功能分析发现主要与跨膜运输、结合、生物合成和细胞器组装等有关,信号途径主要参与核糖体、糖酵解/糖生成等。结论:针刺对脂筏募集蛋白质种类具有良性调节作用,针刺可能通过对特异性蛋白质的募集以达到治疗AD的目的。
Objective:In order to explain acupuncture's possible mechanisms of Alzheimer's disease,we detected SAMP8 hippocampus proteome changes in lipid rafts after acupuncture. Methods : In the study, SAMP8 were divided into control group and acupuncture group, SAMR1 as normal control. The methods which included DTT extraction of hippocampus lipid rafts, HPLC MS / MS shotgun analysis for protein identification, GO and KEGG analysis identified protein functions and signal analysis were used separately. Results :467 kinds of proteins were identified in SAMR1 group,445 kinds in SAMP8 control group. Except of 339 kinds of protein, 119 kinds of proteins only appeared in SAMR1 group, 106 kinds only in SAMP8 control group. After acupuncture treat- ment,92 kinds of proteins of SAMP8 in control group (106) disappeared. 103 kinds of proteins were raised by acupuncture specially,in which 36 kinds were unique to SAMR1. 103 kinds of proteins were functionally analyzed and focused on transmembrane transport, binding, biosynthesis and organelle assembly. Their related signaling pathways mainly included ribosome, glycolysis/gluconeogenesis and so on. Conclusion:Acupuncture has benign regulatory role in lipid rafts raised protein species, and treats AD by raising specific proteins.
