创伤后应激障碍大鼠杏仁核神经元Akt和mTOR磷酸化降低 被引量:2

AKT AND MTOR PHOSPHORYLATION REDUCED IN AMYGDALA OF POSTTRAUMATIC STRESS DISORDER RATS

摘要 目的:探讨创伤后应激障碍(PTSD)大鼠杏仁核Akt和mTOR蛋白表达的变化。方法:随机将40只雄性Wistar大鼠分为对照组和模型组,采用改良的单一连续应激方法(SPS&S)建立PTSD大鼠模型,分别采用免疫组织化学染色和Western blot法检测杏仁核p-Akt和p-mTOR蛋白的表达。结果:模型组大鼠杏仁核p-Akt、p-mTOR的相对表达水平显著低于对照组,组间比较差异具有统计学意义(P<0.05)。结论:PTSD大鼠杏仁核Akt和mTOR磷酸化水平降低,可能与PTSD时杏仁核神经元的凋亡增加有关。 Objective:To observe the changes of Akt and mTOR expression in amygdala of posttraumatic stress disorder(PTSD) rats.Methods:40 male Wistar rats were randomly divided into control group and model group.Single inescapable electric foot shock after SPS procedures(SPS S) was used to made PTSD rats' model.Immunohistochemical staining and Western blot were respectively used to detect the p-Akt and p-mTOR protein expression in amygdala.Results:Compared with control group,the p-Akt and p-mTOR protein expression in amygdala of PTSD model rats reduced obviously(P 0.05).Conclusions:The phosphorylated Akt and mTOR protein expression decreased in amygdala of PTSD rats,which may be concerned with increasing of neurons apoptosis in amygdala during PTSD.
