摘要
研究建立高速逆流色谱法分离制备丹参中丹酚酸B和迷迭香酸.丹参药材经过提取、大孔吸附树脂吸附处理得到粗品,然后以V(正己烷)∶V(乙酸乙酯)∶V(乙醇)∶V(水)=3∶7∶1∶9为两相溶剂体系,上相为固定相,下相为流动相,流速1.8mL/min,检测波长280nm,利用制备型HSCCC分离制备丹参中丹酚酸B与迷迭香酸,利用HPLC测定分离得到的化合物Ⅰ和Ⅱ.纯化后的丹酚酸B(Ⅰ)和迷迭香酸(Ⅱ)经HPLC检测,质量分数分别达到98.4%和95.0%.结果表明:该方法快速简便,制备样品纯度高.
The crude extract of Salvia miltiorrhiza separated through macroporous resin in order to enrich the target compounds preliminary.Then the target compounds separated by high-speed countercurrent chromatography (HSCCC) with a two-phase solvent system contained n-hexane ∶ ethyl acetate ∶ ethanol ∶ water (3 ∶ 7 ∶ 1 ∶ 9,in volume ratio).The upper phase was stationary phase while the lower phase was mobile phase with a flow rate of 1.8 mL/min and the wavelength detection was set at 280 nm.Salvianolic acid B (Ⅰ) and rosmarinic acid (Ⅱ) were obtained and contents were 98.4% and 95.0%,respectively.The results reveal that the method is simple and efficient with a high purity in separation of salvianolic acid B and rosmarinic acid from Salvia miltiorrhiza.
