中药复骨方对糖皮质激素性骨质疏松大鼠骨折愈合的影响

摘要 目的观察中药复骨方对糖皮质激素性骨质疏松(GIOP)大鼠骨折愈合的影响。方法将64只SD大鼠随机均分为4组:基础组、模型组、治疗组和对照组。4组大鼠均建立GIOP骨折动物模型。术后基础组和模型组给予蒸馏水灌胃,治疗组以复骨方药液灌胃给药,对照组以伤科接骨片药液灌胃,持续8周。观察各组股骨骨密度、血清碱性磷酸酶(ALP)和骨钙素(BGP)变化,比较各组骨组织形态计量学参数和生物力学测量结果。结果治疗组、对照组大鼠血清ALP、BGP活性、股骨BMD均高于模型组;模型组大鼠股骨骨小梁面积百分数(TbAr%)、骨小梁数量(TbN)、平均骨小梁厚度(TbTh)均较基础组下降,骨小梁分离度(TbSp)较基础组增高;治疗组、对照组的TbAr%、TbN、TbTh均高于模型组,TbSp低于模型组;治疗组和对照组的最大载荷、最大挠度及最大应力高于模型组;以上各组数据模型组与基础组、治疗组与模型组间各项结果比较差异有显著性(P<0.05),治疗组与对照组各项结果比较差异无显著性(P>0.05)。结论中药复骨方能够提高GIOP骨折大鼠的BMD,升高ALP、BGP活性,改善骨结构,增强骨强度,促进骨生成。复骨方对模型大鼠骨组织计量学和生物力学的影响与伤科接骨片作用相似,其有助于促进GIOP大鼠骨折的愈合。 Objective To observe the effect of Chinese medicine Fugu prescription in treatment of rats with glucocorticoid-induced os-teoporotic( GIOP)fracture. Methods A total of 64 SD rats were randomly divided into 4 groups with 16 rats in each group. All rats had estab-lished bone fracture in the middle of right femora. The oral administration was started at second day of modeling,rats in basic group and model group were given with distilled water 10 ml/kg,rats in treatment group were given with Chinese medicine Fugu prescription,rats in control group were administrated with Shangke Jiegu tablets,once per day for 8 weeks. Serum levels of ALP and BGP were detected by automatic biochemical an-alyzer. BMD was examined by dual-energy X-ray absorptiometry. Rats were sacrificed in 8 weeks after occurring fracture,femurs were removed for biomechanics test and morphological observation. Results The BMD value,serum ALP and BGP activity of femora in rats of treatment group were higher than those of model group. The percentage of trabecular area( TbAr%),the number and thickness of trabecula( TbN,TbTh)in mod-el group were all decreased,but the trabecular bone space( TbSp)was increased;TbAr%,TbN and TbTh in treatment group and control group were higher than those of model group,and TbSp was lower than that of model group. The bone biologic mechanical performance in model group was dropped. The biomechanical analysis showed better results in treatment group than that of control group by three- point bending technique. There was no significant difference between treatment group and control group. Conclusion Chinese medicine Fugu prescription can affect bone metabolism,increase bone formation and improve biomechanical characteristics of rats with femora of GIOP fracture. It has a similar effect with Shangke Jiegu Tablets,and it can contribute to the healing of GIOP fracture.
