鼓槌石斛花总黄酮及挥发性成分研究 被引量:11

Total flavonoids and essential oil of Dendrobium chrysotoxum flower

摘要 研究了滇产鼓槌石斛花中的总黄酮及挥发性成分。分别用超声波法和热回流法提取鼓槌石斛花总黄酮,以芦丁作为标准品,采用紫外可见分光光度法对石斛花中的总黄酮含量进行测定;采用水蒸气蒸馏法提取鼓槌石斛花中的挥发性成分,采用GC-MS技术结合人工解析分析挥发油的化学组成。结果显示,总黄酮在10.08~100.8 mg/L浓度范围内呈现良好线性关系(R2=0.9994),方法在60 min内呈现良好稳定性(RSD=0.19%);2种方法提取的总黄酮含量分别为0.89%和0.27%,高效液相色谱分析显示2种提取方法所得黄酮的组成近似、含量不同,整体而言超声波法提取效率更高;挥发油经GC-MS分析共检出115个峰,鉴定了其中的42个,占挥发油总量的59.1%。 Total flavonoids were extracted from the flowers of Dendrobium chrysotoxum by ultrasonic method and heat refluxing method separately and then measured by ultra violet spectroscopic method with rutin as the standard sample. The essential oil of Dendrobium chrysotoxum was obtained by steam distillation with hexane and analyzed by GC-MS, in which the constituents were identified by the mass spectra, the relative percentage of the oil constituents calculated from the GC peak areas. The result showed that ultrasonic method is more efficient than traditional heat refluxing method, the contents of flavonoids being 0.89% and 0.27% respectively. Differences in compositions can be observed on spectra of higher performance liquid chromatography (HPLC). The determination method showed good liner relationship between 10.08-100.8 mg/L (R2=0.9994) and good stability within 60 min (RSD=0.19%). For the GC-MS study on essential oil, 115 peaks can be observed in which 42 peaks were determined.
