醇提法提取海藻多糖及其α-淀粉酶的抑制活性研究 被引量:9

Extraction of seaweed polysaccharide by ethanol and inhibitory activity of α-amylase

摘要 采用醇提法从海藻中提取多糖,初步纯化,以胰α-淀粉酶为靶酶,4-硝基苯-α-D-吡喃葡萄糖苷(PNPG)为底物,测定其对α-淀粉酶的抑制活性。结果表明,提取率为6.0%;以多糖浓度为12 mg/mL时的抑制率相比较,醇提多糖和脱蛋白醇提多糖的抑制率分别为60.5%和41.35%,完善了海藻多糖的降糖机制;醇提多糖和脱蛋白醇提多糖的IC50分别为2.240 mg/mL和2.630 mg/mL,前者对α-淀粉酶的抑制率高于后者,表明糖蛋白对多糖的降糖活性有协同作用。 The seaweed polysaccharide was extracted by ethanol and preliminary purified. The inhibitory activity of a-amylase was determined in the condition of PNPG as substrate. The results show that the extraction rate was 6.0%. The inhibition rate of seaweed polysaccharide and protein removed polysaccharide were 60.5% and 41.35%, respectively, when polysaccharide concentration was 12 mg/mL, hypoglycemic mechanism of seaweed polysaccharide was replenished. The inhibition rate of seaweed polysaccharide and protein removed polysaccharide IC50 were 2.240 mg/mL and 2.630 mg/mL, the former than the latter, indicated that glycoprotein and polysaccharides had synergistic effect on the hypoglycemic activity.
