家蝇转录组丝氨酸蛋白酶抑制因子分析

Analyses of serine protease inhibitors from the transcriptome of Musca domestica

摘要 目的为了寻求有效控制家蝇的分子靶标,对家蝇丝氨酸蛋白酶抑制因子的主要种类进行初步探讨。方法从家蝇转录组数据库中,筛查出所有注释为丝氨酸蛋白酶抑制因子的Unigenes,利用生物信息学分析技术,对这些Unigenes序列做比对、聚类与结构功能特点分析,并参照已报道的其他物种的丝氨酸蛋白酶抑制因子,对家蝇丝氨酸蛋白酶抑制因子进行初步分类。结果从家蝇转录组库中共筛选出133条注释为丝氨酸蛋白酶抑制因子的Unigenes,其中,值得分析的序列有59条,具有典型丝氨酸蛋白酶抑制因子结构域的有22条,它们被初步分成11类。结论本研究首次鉴定了11种家蝇丝氨酸蛋白酶抑制因子基因,为生物控制家蝇提供了重要的候选靶标。 Objective In order to seek the molecular targets for effectively controlling Musca domestica, the serine protease inhibitors of housefly were classified prehminarily. Methods Based on the functional annotation of uni- genes in M. domestica transcriptomic database, the target unigenes annotated as serine protease inhibitors were screened ont. Their alignment, clustering, characteristics analysis in structure and function were finished by bioinfor- matics analysis technology. Serine protease inhibitors of M. domestica were clearly stated with reference to the type of serine protease inhibitors of other species. Results 133 unigenes annotated as serine protease inhibitors were fil- tered out from M. domestica transcriptomic database. 59 unigenes were valuable to be further analyzed, and 22 uni- genes with the typical dormain of serpin were preliminarily divided into 11 categories. Conclusion This is the first report that M. domestica may have 11 serine protease inhibitor genes and that they may be important candidate targets for biological controlling houseflies.
