摘要
目的:测定不同溶剂对金荞麦中有效成分总黄酮类物质的提取效率,制定简便的金荞麦制剂的制备工艺。方法:采用紫外分光光度法,设定检测波长为510 nm,以芦丁为对照品绘制标准曲线,测定金荞麦中总黄酮含量。结果:在10倍量的溶剂提取下,60%乙醇的金荞麦提取效率最高;芦丁对照品在16~160(μg·mL^-1)呈良好线性关系,求得直线回归方程为Y=0.000 88X+0.160 2,r=0.997 4;用60%的乙醇为提取溶剂提取金荞麦中总黄酮的含量最高;精密度试验的RSD为0.56%;重复性试验的平均值为2.76%,RSD=2.92%(n=6);加样回收率试验的平均值为98.43%,RSD=1.78%。结论:以60%的乙醇为溶媒提取金荞麦中的总黄酮类成分提取率最高,稳定性好,方法简便,适用于我院金荞麦制剂的生产。
Objective:To determinate the efficiency extracting the total flavonoids from Fagopyrum dibotrys with different solvents,in order to make the preparations to play a better role in treatment and provide a easy preparation process for fago mixture in hospital.Methods:By UV spectrophotometry,setting the detection wavelength of 510 nm and taking Rutin as a reference so as to make the standard curve,the content of total flavonoids in fagopyrum dibotrys was determined.Results : In extraction with 10 times of solvent,the efficiency extracting the total flavonids from Fagopyrum dibotrys with 60% ethanol was highest; the reference Rutin showed a good linear relationship at 16~ 160(μg·mL^-1),the obtained linear regression equation was Y =0.000 88 X + 0.160 2,r =0.9974; the content of total flavonoids extrated from fago with 60% ethanol as extraction solvent was highest; RSD of precision test was 0.56%; the average value of repeatability test was 2.76%,RSD=2.92%(n=6); the average value of recovery rate test for added sample was 98.43%,RSD=1.78%.Conclusions: The efficiency extrating the total flavonoids from Fago with 60% ethanol as extraction solvent is highest,its stability is better,the method is easy,and suitable for the production of Fago Mixture preparations in our hospital.
