Study on quality of Eucommiae Cortex using flow injection-mass spectrometric fingerprinting method combined with chemical pattern recognition approaches

摘要 目的利用直接进样-质谱检测指纹图谱技术结合化学模式识别法研究不同产地杜仲药材的质量。方法样品经简单提取后,不经色谱分离,直接进入质谱仪进行分析,产生的m/z 100~1 000的离子响应强度信息经预处理后由分析软件Simca-P进行主成分分析(PCA)和偏最小二乘法判别分析(PLS-DA)。结果经PCA和PLS-DA分析得到的分布图将来源于不同产地的杜仲聚为4类,反应了其化学成分的差异性;通过载荷图则可发现在分类过程中起决定作用的化学标记物。结论该方法作为一种极具特点的指纹图谱分析技术,可以被广泛用于药材的真伪鉴别、基源鉴定及不同产地药材的快速判别研究。 Objective To study the quality of Eucommiae Cortex from different resources using flow injection mass spectrometric fingerprinting (FIMS) method combined with chemical pattern recognition approaches. Methods Different Eucommiae Cortex samples were extracted in a simple way and were directly injected into the mass spectrometer for the analyses with no column used. Based on the intensities of the ions at rn/z 100--1 000, principal components analysis and partial least square discrimination analysis were performed using Simca-P software. Results Eucommiae Cortex samples from different resources were classified into four clusters in scores plot, and the biomarkers which played the most important roles in classification were screened out. Conclusion As a novel and characteristic fingerprinting method, FIMS can b^e widely and efficiently used in true or false identification, identification of origins, and rapid discrimination of herbs from different regions.
