肉桂醛对黑色素瘤小鼠的抑瘤作用及其机制研究 被引量:8

摘要 目的研究肉桂醛对黑色素瘤的影响及其机制。方法取20只Balb/c null雌性裸鼠建立黑色素瘤模型,分为生理盐水组和肉桂醛组,每组10只,第2周开始每鼠分别注射3 ml生理盐水(生理盐水组)和3 ml、10μg/ml肉桂醛药物溶液(肉桂醛组),比较两组小鼠瘤体体积差异,并通过免疫组化法观察两组之间的新生血管数量,免疫印迹法检测血管内皮生长因子(VEGF)和缺氧诱导因子(HIF)-α的表达水平。结果第3周肉桂醛组的黑色素瘤瘤体体积和新生血管数量与生理盐水组比较,均有显著下降(P<0.05),在肉桂醛组中的HIF-α和VEGF表达也显著低于生理盐水组(P<0.05)。结论肉桂醛通过抑制VEGF和HIF-α的表达,进而抑制黑色素瘤新生血管的生成,进而抑制黑色素瘤的生长。 Objective To study the effect and its mechanism of Cinnamic Aldehyde on melanoma. Methods Twenty Balb/c null female nude mice were selected to establish melanoma model, and then were divided into physiologi-cal saline group (n=10) and Cinnamic Aldehyde group (n=10). In the second week, each mouse in physiological sa-line group was injected with 3 ml physiological saline, while each mouse in Cinnamic Aldehyde group was injected with 3 ml 10 μg/ml Cinnamic Aldehyde drug solution. The difference in tumor volume of mice between the two groups was com-pared, and the neovessels number of the two groups was observed by immunohistochemistry, and expression levels of vas-cular endothelial growth factor ( VEGF ) and hypoxia-inducible factor-α ( HIF-α) were detected by Western Blotting method. Results In the third week, the values of melanoma tumor volume and the neovessels number in Cinnamic Alde-hyde group were significantly decreased, compared with those in physiological saline group (P〈0. 05), and expression levels of HIF-α and VEGF in Cinnamic Aldehyde group were also significantly lower than those in physiological saline group (P〈0. 05). Conclusion The Cinnamic Aldehyde can inhibit the neovascular growth of melanoma by inhibiting the expressions of VEGF and HIF-α to inhibit the melanoma growth.
