摘要
目的比较肝癌组织、癌旁组织及正常肝组织中叉状头状转录因子(forkhead transcription factor,FoxP3)的表达,探索FoxP3基因在肝癌细胞免疫逃避中的作用。方法采集15例肝癌肝移植手术全肝切除标本,实时荧光RT-PCR检测并比较肝癌组织、癌旁组织及正常肝组织中FoxP3 mRNA。结果实时荧光定量RT-PCR各组标本FoxP3 mRNA的结果,肝癌组织(A组):肝癌组织为27.5339±2.1647;癌旁组织(B组):癌旁组织为24.9804±2.35755;正常肝组织(C组):为24.3800±2.24896)。可见A组FoxP3 mRNA含量最高,明显高于B组和正常肝组织C组,差异有统计学意义(P<0.01);癌旁组织B组FoxP3 mRNA含量略高于正常肝组织C组,差异无统计学意义(P>0.05)。结论在肝癌局部表达FoxP3的Treg增多,形成局部免疫耐受,可能是肝癌复发转移的主要原因。
Objective To explore the role of forkhead transcription factor(FoxP3) in metastasis of HCC. Methods The fresh hepatocellar carcinoma tissue(Group A), paracarcinoma tissue(Group B) and control liver tissue(Group C) were taken from 15 hepatocellar carcinoma patients. Then the FoxP3 mRNA in these tissues was detected using real-time RT-PCR respectively.Results In hepatocellular carcinoma tissue(Group A),the expression of FoxP3 mRNA was 27.5339 ± 2.16474;in paracarcinoma tissue(Group B) the expression of FoxP3 mRNA was 24.9804 ± 2.35755 and in control group(Group C) it was 24.3800 ± 2.24896.Expression of FoxP3 mRNA was increased in hepatocellular carcinoma tissue compared to group B or group C, respectively(P〈 0.01);However in paracarcinoma tissue(Group B) expression of FoxP3 mRNA was not up-regulated compared to Group C(P〉 0.05).Conclusion Up-regulated expression of FoxP3+Treg induced by HCC is possibly responsible for the immunotolerance in HCC reoccurrence and metastasis.
