摘要
目的:探讨益气补肾法对自然流产模型小鼠脾脏CD4+CD25+T细胞上Foxp3因子的表达及妊娠结局的影响。方法:将雌性CBA/J与雄性DBA/2小鼠合笼交配,造成自然流产模型。将CBA/J孕鼠随机分成5组,中药高剂量组、中药中剂量组、中药低剂量组、阳性对照组(CsA组)、阴性对照组(NS组),分别于妊娠第9天、第14天将其处死,其中孕第14天计算胚胎吸收率,并采用流式细胞术分析Foxp3因子比例。结果:1各种干预手段对自然流产模型妊娠结局的影响:孕第14天,各治疗组与阴性对照组相比,仅中药中剂量组与其相比差异存在统计学意义(P<0.05);中药各治疗组与阳性对照组相比不存在统计学差异;中药中剂量组与中药低剂量组相比存在统计差异意义(P<0.05)。2各种干预手段对自然流产模型小鼠脾脏CD4+CD25+T细胞上Foxp3因子表达的影响:孕第9天,各治疗组与阴性对照组相比,差异均存在统计学意义(P<0.05,P<0.01);各治疗组与阳性对照组相比,仅中药高剂量组与其相比差异存在统计学意义(P<0.01);中药高剂量组与中药中剂量组、中药低剂量组相比差异存在统计学意义(P<0.05)。孕第14天,各组间相比,仅中药中剂量组与阴性对照组相比差异存在统计学意义(P<0.05)。结论:益气补肾法可使自然流产模型小鼠脾脏CD4+CD25+T细胞上Foxp3因子高表达并有降低胚胎吸收率的趋势,为益气补肾中药在临床的应用提供了一定的理论依据。
Objective: To study the effect of nourishing kidney and qi therapy on the expression of Foxp3 in spleen of spontaneous abortion mice and the influence on pregnancy outcome. Methods: CBA/J female mice were mated with DBA/2 male mice in order to establish spontaneous abortion model, which were randomly divided into 5 groups, the high, medium and lowdose Chinese medicine groups, the positive control group(treated with CsA), the negative control group(fed with normal saline), then these mice were sacrificed on the 9th and 14 th day of gestation respectively. The embryo absorption rate was counted on the 14 th day of gestation, and proportion of Foxp3 factor were analyzed by flow cytometry. Results: 1The effect of different interventions on pregnancy outcome of spontaneous abortion model: Compared with the negative control group, only the mediumdose Chinese medicine group had significant statistically differences on the 14 th day of gestation(P〈0.05). Compared with the positive control group, all Chinese medicine treatment groups had no significant difference. The medium and low-dose Chinese medicine groups showed significant differences(P〈0.05), and there was no difference among all the treatment groups. 2The effect of different interventions on the expression of Foxp3 factor of CD4+CD25+ on T cells in spleen of spontaneous abortion mice: Compared with the negative control group, all treatment groups had significant differences on the 9th day of gestation(P〈0.05, P〈0.01). Compared with the positive control group, only the high-dose Chinese medicine group had significant differences(P〈0.01). The high-dose Chinese medicine group had significant differences compared with the medium and low-dose Chinese medicine groups(P〈0.05), and there was no difference among all treatment groups. Compared with the negative control group,only the medium-dose Chinese medicine group had significant differences on the 14 th day of gestation(P〈0.05). Compared with the positive control group, there was no statistically difference in each treatment group. There was no statistically difference among all the treatment groups. Conclusion: The nourishing kidney and qi therapy can improve the expression of Foxp3 factor of CD4+CD25+ on T cells and reduce the embryo absorption rate in spleen of spontaneous abortion mice model, which can provide a certain theoretical basis for the application of Chinese drugs for nourishing kidney and qi in the clinical practice.
