宫内铅暴露脐带血DNA甲基化450K芯片检测结果的初步分析 被引量:1

摘要 目的:研究宫内铅暴露新生儿脐带血中DNA甲基化450K芯片检测结果的差异情况。方法:从采集的脐带血样本中,按铅暴露含量(均数)分为高(G1)、中(G2)、低(G3)暴露组,每组4例;用Illumina Infinium Human Methylation 450 BeadArray芯片,在单核苷酸水平上评估脐带血DNA中约485 000个CpG位点的甲基化情况,并对甲基化表达差异基因进行生物信息学分析。结果:G1与G2、G3组和G2与G3组CpG甲基化位点差异为:306个(208个高位点,98个低位点)、227个(111个高位点,116个低位点)和237个(91个高位点,146个低位点);各组两样本基因TSS1500、TSS200、5’UTR、1stExon、Gene body和3’UTR内差异甲基化区域个数为:4、5、3、6、5、8,2、3、2、5、1、7与4、5、2、1、5、6;CpG Island、N shelf、N Shore、S shore、S Shelf内差异甲基化区域个数为:10、12、8、10、11,8、14、3、4、12与8、12、6、6、13。经DAVID Bioinformatics Resources 6.7(Bioinformatics database)检索,确定这些差异CpG位点所在基因和差异甲基化区域身份和功能,这些基因位点和区域主要与细胞形态发生、Ca2+运输通道、细胞黏附、细胞凋亡,以及与抗原的发生和提呈、免疫应答调节等密切相关。结论:铅宫内暴露可能通过基因组的表观遗传调控而影响胎儿的健康发育,为下一步的验证提供了依据。
