摘要
目的:探讨成年慢性应激大鼠自发活动及海马区脑源性神经营养因子(BDNF)的表达。方法:成年雄性SD大鼠随机分为正常对照组和应激造模组,每组各6只,经28 d慢性不可预测温和应激(CUMS),以开场实验检测大鼠自发活动,Western blot检测海马BDNF表达变化。结果:28 d CUMS后,应激造模组总路程((5.3±1.5)m)、中央路程((0.5±0.3)m)、周边路程((4.8±1.5)m)均明显少于正常对照组((19.4±2.5)m),((1.8±0.3)m),((17.7±2.6)m)(P<0.001);应激造模组海马区BDNF表达水平(1.0±0.4)与正常对照组(0.8±0.3)比无统计学意义。结论:慢性应激能致动物抑郁样行为,对大鼠海马区BDNF表达无明显影响。
Objective:To investigate the locomotor activity and brain-derived neurotrophic factor(BDNF)level in the hippocampus of adult rats subjected to chronic stress. Methods:Adult male SD rats were divided randomly into normal control group(n=6)and stress-model group(n=6). After 28 d chronic unpredictable mild stress(CUMS),the locomotor activity in rats was detected by open field test. The changes of BDNF level were detected in the hippocampus by Western blot. Results : After 28 d CUMS, the total distance((5.3±1.5)m), middle distance((0.5±0.3)m)and border distance((4.8±1.5)m)in the stress-model group were significantly less than those in the control group((19.4±2.5)m,(1.8±0.3)m,(17.7±2.6)m)respectively(P0.001). There was no significant difference in the expression of BDNF in the hippocampus between stress-model group(1.0±0.4)and normal control group(0.8±0.3). Conclusion:The chronic stress can result in depressive-like behaviors,and make no difference to the expression of BDNF in the hippocampus of rats.
