摘要
目的:观察补肾健脾方药对骨质疏松症小鼠骨骼、骨骼肌异柠檬酸脱氢酶(IDH)含量变化的影响,评价中医防治骨质疏松症的效率。方法:将OPG基因敲除小鼠随机分为模型对照组、补肾组、健脾组和福善美组,用野生型小鼠作为正常对照组。Elisa法测定小鼠骨骼、骨骼肌IDH含量。结果:1各组小鼠较正常组骨骼和骨骼肌中IDH表达显著降低(P<0.01);2补肾组、福善美组较模型对照组小鼠骨骼和骨骼肌中IDH表达明显升高(P<0.01);3健脾组较模型对照组小鼠骨骼和骨骼肌中IDH的表达有升高,但差异无统计学意义。结论:1骨质疏松症的形成,骨骼与骨骼肌功能下降,局部能量代谢减低,导致IDH含量异常变化。2补肾、健脾方法通过提高骨质疏松症大鼠的骨骼、骨骼肌IDH的含量,对骨质疏松症具有一定的防治作用。
Objective: To compare the effect of invigorating kidney,invigorating spleen in treating osteoporosis by investigating expression of IDH in the skeleton and skeletal muscle of osteoporosis mice. Method: Except the normal control group,OPG knockout mice were taken as the osteoporosis model. OPG-/- mice were randomly divided into model control group( no herb inducted),invigorating kidney group,invigorating spleen group and Fosamax group. Euzymelinked immunosorbent assay were taken to analyze the content of IDH in skeletal and muscle. Result: All the OPG-/-mice shows significantly lower IDH in the muscle than normal control group. Invigorating kidney dramatically reverse the decrease of IDH expression( P〈0. 01),and also Fosamax( P〈0. 01). However,the invigorating spleen group doesn't show statistically significant change. Similar result turns out in skeletal. Conclusion: 1Osteoporosis causes the failure of bone and muscle in function,which is marked with the lower IDH expression in parallel to the energetic metabolism. 2 Treating with invigorating the kidney and invigorating the spleen can partially reverse IDH expression in the skeleton and skeletal muscle,which indicates that it can efficiently prevent and cure osteoporosis.
