电针对慢性应激抑郁模型大鼠松果体影响的研究 被引量:1

Effects of electroacupuncture on pineal body in rats with chronic stress depression

摘要 目的观察电针针刺"百会""印堂"对慢性应激抑郁模型大鼠松果体的影响,探讨电针抗抑郁的作用机制。方法将24只SD雄性大鼠随机分为3组:空白组、模型组、电针治疗组。抑郁症模型选用慢性轻度不可预见性应激和孤养方法复制,通过开野实验观测大鼠的行为学变化,应用透射电子显微镜观察松果体的超微结构变化。结果与空白组比较,模型组的水平穿越格数、直立运动次数明显减少(P<0.05);与模型组比较,电针治疗组的水平穿越格数、直立运动次数显著增多(P<0.05);与空白组比较,模型组大鼠松果体细胞数目减少,排列疏松,线粒体减少,呈不同程度肿胀,髓鞘板层分离、变性,出现凋亡样变化。电针治疗组松果体数目增多,排列紧凑,核仁明显,髓鞘尚规整、致密,线粒体肿胀减轻。结论电针可以增加慢性应激模型大鼠松果体细胞数目,减少髓鞘脱失,保存线粒体功能,可能是电针抗抑郁作用的机制之一。 Objective To observe the effects of electroacupuncture( EA) at Baihui( GV 20) and Yintang( GV 29) on pineal body in rats with chronic stress depression,so as to explore the action mechanism of EA on antidepression. Methods Twenty-four SD male rats were randomly divided into 3 groups: a blank group,a model group and an EA group. The depression model of rats was established by chronic unpredictable mild stress and isolation raising method; open-field test was used to detect the behavior change of rats;transmission electron microscopy( TEM) was used to observe the ultrastructural changes of pineal body. Results Compared with the blank group,the number of horizontal across and the frequency of vertical movement in the model group were significantly reduced( P 〈0. 05); compared with the model group,the number of horizontal across and the frequency of vertical movement in the EA group were significantly reduced( P 〈0. 05). Compared with the blank group,the number of pineal body in the model group was reduced with loose arrangement,and mitochondrion was decreased with swelling,and myelin sheath had lamellar separation and degeneration,even apoptosis. The number of pineal body in the EA group was increased with tight arrangement,and nucleolus was obvious,and myelin sheath was structured and compact,and the mitochondrion was less swelling. Conclusion Electroacupuncture can increase number of pineal body in rats with chronic stress depression,reduce the loss of myelin sheath,protect the function of mitochondrion,which may be one of the action mechanisms of electroacupuncture on antidepressant.
