海珠区集中空调冷却塔水嗜肺军团菌及rpoB基因分型分析

Analysis of strain type of legionella pneumophila and molecular genotyping of rpoB sequence from cooling water in central air conditioning system in Haizhu district of Guangzhou

摘要 目的了解广州市海珠区公共场所集中空调冷却塔水的嗜肺军团菌污染状况。方法随机抽样采集36家公共场所集中空调冷却塔水85份,通过血清学凝集试验、实时荧光PCR技术对嗜肺军团菌菌株进行鉴定及rpoB基因测序分型分析。结果采集集中空调冷却塔水85份,分离得到嗜肺军团菌23株,阳性率27.1%,血清型别多样,以LP1和LP6型为主,分别占检出株数的34.8%(8/23)和26.1%(6/23),以rpoB基因序列分型得到种属鉴定结果。结论广州市海珠区集中空调系统嗜肺军团菌污染较严重,血清型别多样化,rpoB基因分型具有地区特异性。 Objective To understand contamination of Legionella pneumophila in cooling tower water of central air conditioning in the public places. Methods Eighty-five samples of cooling water in central air conditioning system in 36 public places of Haizhu, Guangzhou. The bacteriologic examination and genotyping were performed by seroagglutination test and PCR test. Results A total of 23 strains of legionella pneumophila were isolated from the 85 samples. The positive rate was 27.1% and the serum type was diverse. LP1 and LP6 are dominated with the rate of 34.8% (8/23),26.1% (6/23).The identification of Legionella pneumophila species was done by rpoB gene sequence typing. Conclusion The Legionella pneumophita contamination was serious in central air conditioning system in Guangzhou of Haizhu. The serum type was diverse and the rpoB gene was with regional specific.
