正天丸对偏头痛大鼠的血浆代谢组学研究 被引量:7

摘要 [目的]运用代谢组学方法,研究正天丸治疗偏头痛的机制。[方法]给药和造模后取大鼠血浆,通过超高效液相色谱结合高分辨率飞行时间质谱联用(UPLC-Q-TOF/MS)技术对各组血浆进行代谢谱检测。采用Micromass Markerlynx软件、SIMCA-P+软件进行多维统计分析,通过偏最小二乘法一判别分析(PLS-DA)进行模式识别。[结果]色谱图显示正天丸组离子峰较模型组明显减少。散点分布图提示正天丸组和模型组聚类明显。荷载图外周散在点提示有多个化合物对两组的聚类起了重要作用。初步推断3个化合物(磷脂酰胆碱、3,4-二羟苯乙二醇以及未确定化合物)在分类中起了重要作用。[结论]本研究建立了基于大鼠血浆的UPLC-Q-TOF/MS代谢组学研究方法,初步阐释了正天丸通过纠正神经细胞信息传递紊乱和提高疼痛阈值治疗偏头痛的代谢组学机制。 [Objective]To investigate the metabonomic of therapeutical effect of Zhengtian Pills to the migraine in rats.[Methods] Blood serum from migraine rats was taken and UPLC-Q-TOF/MS was used to detect the metabolite profiles of the blood serum samples of every groups.The primary data were pretreated by the Markerlynx software.Then the data were analyzed by pattern recognition method which was composed of PLS-DA in SIMCAP+.[Results] The chromatogram showed that the molecular ion of zhengtian pill had significandy reduced compared with model group.The splashes distribution graph showed that,the cluster in Zhengtian Pill group and model group was both obvious.Load diagram peripherypoints indicated that,there was a lot of chemical compounds playing an important role on the cluster of the two groups.Three potential biomarkers were identified,which played an important role on the cluster of the two groups.[Conclusion] The topic established a method of metabonomic analysis for blood serum from rats using LC/MS.The study showed the mechanism of metabolism for the therapy of migraine with Zhengtian Pills.The results will be great helpful to explain the theory of Chinese patent medicine using biological science of modem time.
