杜仲饮片中京尼平苷酸和京尼平苷的含量测定 被引量:6

摘要 目的:建立中药杜仲中京尼平苷酸(geniposidic acid,GPA)和京尼平苷(geniposide,GP)含量的HPLC测定法。方法:应用HPLC对盐炙杜仲饮片中的GPA和GP进行测定,色谱柱为Agilent TC-C18柱(250 mm×4.6 mm,5μm),柱温为30℃,以甲醇-0.1%磷酸(25∶75,v/v)为流动相,流速为1.0 mL/min,检测波长为240 nm。结果:GPA线性回归方程为YGPA=24.701ρ+39.014(r2=0.999 8),在进样浓度5.12~500μg/mL范闱内呈良好的线性关系;GP线性回归方程为YGP=26.661ρ-25.017(r2=0.999 7),在进样浓度4.506~440μg/mL范围内呈良好的线性关系;GPA和GP加标回收率分别为100.20%和99.80%,其RSD分别为2.15%和2.26%。结论:本法操作简便可行,重复性较好,适用于杜仲药材中京尼平苷酸和京尼平苷的含量测定,及对其药材质量优劣的评价。 Objective: To develop a HPLC method for the determination of geniposidic acid and geniposide in Eucommia ulmoides Oliv.. Methods: An Agi|ent TC C18 column (250 mm×4.6 mm, 5μm) was used at 30 ℃with the mobile phase of methanol-0.1% phosphatic acid with volume ratio of 25:75. The flow rate was set at 1.0 mL/min, and the detection wavelength was 240 nm. Results: The linear regression equations of G PA and GP were respectively YGPA = 24.701 ρ+39.014(r^2= 0.999 8) and YGP= 26.661 ρ - 25.017 (r^2= 0.999 7), and linear of two concentratious were between 5.12 and 500 μg/mL, and were between 4.506 and 440 μg/mL, respectively. Conclusion: The method is quick, simple and repeatable for simultaneous determination of geniposidic acid and geniposide in Eucommia ulmoides Oliv., and it can be used for the intergrated analysis of the two biologically active components.
