刀豆素A诱导人乳腺癌细胞自噬性死亡及其机制

摘要 目的探讨刀豆素A对人乳腺癌细胞MCF-7的杀伤作用及其机制。方法采用MTT法检测不同浓度ConA(concanavalin A)处理MCF-7细胞后,MCF-7细胞的增殖情况,并检测自噬标记蛋白LC-3Ⅱ,加入自噬特异性抑制剂3-甲基腺嘌呤(3-MA)后再检测MCF-7细胞对ConA的敏感性。利用基因沉默技术,抑制BNIP3的表达,再检测ConA处理后MCF-7细胞的增殖和自体吞噬。结果 ConA可显著抑制MCF-7细胞的增殖(P<0.01),并诱导其发生自噬性死亡。ConA处理后MCF-7细胞的BNIP3显著增高(P<0.05,P<0.01),利用小RNA抑制BNIP3的表达后,ConA对MCF-7的增殖抑制作用降低(P<0.01),并抑制了ConA诱导的自体吞噬效应。结论刀豆素A通过上调BNIP3的表达诱导人乳腺癌细胞发生自噬性死亡。 Objective To investigate the effect of concanavalin A on proliferation of MCF-7 cells and the un-derlying mechanism. Methods MCF-7 cells were treated with different concentrations of concanavalin A and then the proliferation of MCF-7 was detected by MTT method and LC-3II was labeled. 3-MA was added to detect the sensitivity of MCF-7 cells to concanavalin A. After silencing the expression of BNIP3 with small RNA,the prolifer-ation and autophagy of MCF-7 cells treated with concanavalin A were detected. Results Concanavalin A signifi cantly inhibited the proliferation of MCF-7 cells(P〈0.01) and induced autophagic cell death. The expression of BNIP3 significantly increased in MCF-7 cells when treated with ConA(P〈0.05,P〈0.01). After silencing BNIP3 with small RNA,the effect of inhibition and autophagy of concanavalin A decreased in MCF-7 cells. Conclusion Con-canavalin A can induce autophagic cell death of MCF-7 cells via upregulating the expression of BNIP3.
