摘要
目的观察不同比例黄连配伍肉桂对2型糖尿病大鼠糖脂代谢及氧化应激的影响。方法采用小剂量链脲佐菌素尾静脉注射加高脂高热卡饲料喂养的方法建立大鼠2型糖尿病模型,将大鼠随机分为模型组、肉桂组、交泰丸1组、交泰丸2组,各治疗组分别予以相应药物灌胃干预,另设正常对照组。干预10周后比较各组大鼠体质量、空腹血糖(FBG)、口服葡萄糖耐量试验(OGTT)、空腹血清胰岛素(FINS)以及血脂水平,并检测血清中丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活性以观察药物对大鼠氧化及抗氧化作用的影响。结果与正常组比较,模型组大鼠OGTT异常,FBG、FINS升高,血脂紊乱,MDA活性升高,SOD及GSH-PX活性下降;与模型组比较,各药物干预组FBG及FINS降低,OGTT及血脂紊乱改善,MDA活性下降,SOD及GSH-PX活性升高,其中交泰丸2组降糖调脂及抗氧化应激效果最优。结论不同比例黄连配伍肉桂对2型糖尿病大鼠糖脂代谢及抗氧化应激的影响不同,其机制可能与黄连肉桂两药配伍后影响各自降糖成分的溶出及代谢等有关。
Objective: To explore the effects of different compatibility ratio of Cortex Cinnamomi and Rhizoma Coptidis on blood glucolipid metabolism and oxidative stress in type 2 diabetic rats. Methods: The model of type 2 diabetic rats was established by injecting streptozotocin from tail vein and by feeding with high fat and high caloric diet. Diabetic rats were randomly divided into model group,cinnamon group,Jiaotai pill 1 group (Coptidis Rhizoma:Cortex cinnamomi =4:1 ) and Jiaotai pill 2 group (Coptidis Rhizoma:Cortex cinnamomi= 2:1 ). Rats in dif- ferent treatment groups were given corresponding therapy. Meanwhile normal control group was set. Fasting blood glucose (FBG), fasting blood insulin (FINS), oral glucose tolerance test (OGTT), blood lipid level, malonaldehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) in the serum were determined after ten weeks. The effects on the oxidative stress of rats were observed. Results: OGTY was abnormal,and FBG and FINS increased in rats of model group compared with those in normal control group. Plasma levels were abnormal in model group. Compared With normal group,model group showed higher MDA level and decreased SOD and GSH-PX activity in serum, In cinnamon,Jiaotai pill 1 and Jiaotai pill 2 groups,the SOD and GSH-PX elevated drastically while FBG,FINS,and MDA level decreased compared with model group. Among the three treatment groups,the best effect of hypoglycemic lipid-lowering and anti-oxidant stress occurred in Jiaotai pill 2 group. Conclusion: There are differences in the effects of different compatibility ratio of Cortex Cinnamomi and Rhizoma Coptidis on blood glucolipid metabolism and oxidative stress in type 2 diabetic rats. The potential mechanisms may be related with the dissolution and metabolism of each hypoglycemic element after the compatibility of Cortex cinnamomi and Rhizoma coptidis.
