摘要
目的观察黄连阿胶汤对失眠模型大鼠Th1/Th2平衡的影响。方法将36只SD大鼠随机分为3组,PCPA法复制失眠大鼠模型,ELISA法检测大鼠血清IFN-γ、IL-1β、IL-4、IL-10及IFN-γ/IL-4的水平。结果模型组大鼠IFN-γ、IL-1β的水平明显下降(P<0.01),IL-4、IL-10的表达则明显升高(P<0.01),IFN-γ/IL-4比值降低(P<0.01);黄连阿胶汤组IFN-γ、IL-1β的表达有不同程度的升高(P<0.05,P<0.01),IL-4、IL-10的表达降低(P<0.05),IFN-γ/IL-4比值升高(P<0.01)。结论黄连阿胶汤能提高失眠大鼠Th1细胞因子的表达,降低Th2细胞因子的表达,促进Th1/Th2平衡向Th1方向偏移,是其治疗失眠的可能机制之一。
Objective To observe the effect of Huanglian Ejiao Decoction on serum Th1/Th2 balance of insomnia rats. Methods 36 SD rats were randomly divided into 3 groups. The model of insomnia rats was established by PCPA method. The level of IFN-γ, IL-1β, IL-4, IL-10 and IFN-γ/IL-4 in rats serum were determined with enzyme linked immunosordent as- sa (ELISA) method at the end of the experiment. Results The level of IFN-γ, IL-1β reduced obviously (P 〈 0.01, the expression of IL-4 and IL-10 obviously increased( P 〈 0.01, and the ratio of IFN-γ/IL-4 decreased in the model group; The expression of IFN-γ, IL- 1β increased in different degrees ( P 〈 0.5, P 〈 0.01 ), the expression of IL-4, IL- 10 reduced ( P 〈 0.05), the ratio of IFN-γ/IL-4 increased ( P 〈 0.01 ) in Huanglian Ejiao Decoction group. Conclusion Huanglian Ejiao Decoction can increase the expression of serum Thl cytokines and decrease the expression of serum Th2 cytokines in insom- nia rats. The possible mechanism of Huanglian Ejiao Decoction in the treatment of insomnia may be achieved by promoting the balance of the Th1/Th2 move to Thl direction.
