摘要
目的研究中药御糖丸对糖尿病大鼠心肌能量代谢酶、过氧化物酶体增殖剂活化受体α(PPARα)mRNA表达的影响,探讨该药物防治糖尿病心肌病发生的疗效及可能机制。方法采用SD大鼠腹腔注射STZ方法建立实验动物模型,成模后将大鼠模型分为模型组、御糖丸高、中、低剂量组,造模后8周分别按实验要求灌胃治疗,正常对照组灌生理盐水,连续4周。高效液相色谱法测定大鼠心肌组织肌酸激酶(CK)、乳酸脱氢酶(LDH)和琥珀酸脱氢酶(SDH)活性,逆转录聚合酶链式反应(RT-PCR)测定心肌组织PPAR mRNA表达。结果空白模型组大鼠心肌CK、LDH、SDH活性普遍降低,指标明显低于正常对照组(P<0.01),御糖丸各剂量组指标较空白模型组均有提升,但中剂量组和高剂量组提高显著(P<0.05)。空白模型组PPAR mRNA表达水平显著低于正常对照组(P<0.01),御糖丸各剂量组PPARa mRNA表达水平均高于空白模型组(P<0.05),但中高剂量组提高较为显著(P<0.01)。结论中成药御糖丸能够提升糖尿病大鼠心肌酶CK,LDH,SDH的活性和上调PPARa mRNA表达水平,对改善心肌能量代谢和保护心肌组织具有良好作用。
Objective To study the effect of Yutang Pill on myocardial energy metabolism enzyme agent and peroxidase proliferation activated receptorα (PPARα) mRNA expression of diabetic rats, and to discuss the curative effect and possible mechanism of the drug's prevention and treatment for diabetic cardiomyopathy. Methods Researchers use STZ intraperitoneal injection for SD rats to establish the experimental animal models and then divide the rats into model group, yu-tang pill high dose group, Yutang Pill medium and low dose groups. 8 weeks after the modeling, different lavage treatment, as required above, are applied to different groups for four weeks and control group use saline. High performance liquid chromatography is used to test creatine kinase (CK), lactate dehydrogenase (LDH), succinate dehydrogenase (SDH) activity and reverse transcriptase-polymerase chain reaction (RT-PCR) is used to test the expression of PPARa mRNA. Results Myocardial CK, LDH, SDH activity in blank model group are generally declined and the index is significantly lower than that of the normal control group ( P 〈 0.01 ). For group with different dose of Yutang Pill, the index is higher than the blank model group, among which the medium and high group improved significantly ( P 〈 0.05). PPARαmRNA expression level of blank model group is significantly lower than normal control group ( P 〈 0.01), and PPARαnRNA expression level of group with different dose of Yutang Pill are hizher than blank model group (P 〈 0.05). among which the medium and high group ira-proved significantly ( P 〈 0.01 ). Conclusion Yutang Pill can improve diabetic rats' myocardial enzyme CK, LDH and the activity of SDH and raised PPARαmRNA expression level, which is beneficial to improve myocardial energy metabolism and protect myocardial tissue.
